摘要
目的探讨受体耐受性树突状细胞(DC)在诱导供体特异性免疫耐受中的作用及其可能机制。方法通过以B7反义肽封闭的负载供体抗原的受体DC对受体小鼠进行预处理,3d或2个月取脾脏分离T细胞,与负载供体抗原或无关供体抗原的DC作混合淋巴细胞反应,并且在3d或2个月时以负载供体抗原的受体DC对受体小鼠进行再次免疫后,RT-PCR半定量法测定脾脏中细胞因子mRNA表达。结果以B7反义肽封闭的负载供体抗原的受体DC预处理3d或2个月后的受体T细胞,产生对间接途径提呈的供体抗原的免疫低反应;同时脾脏IL-10表达升高,而IL-2、IFN-γ表达降低。结论 B7反义肽封闭的受体DC可能通过免疫偏移诱导针对间接途径提呈的供体抗原特异性的免疫低反应。
Objective To investigate the effect and possible mechanism of the recipient-tolerogenic dentritic cells(DCs) on the donor-specific immune hyporesponsiveness. Methods The recipient rats were pretreated with the donor-pulsed recipient DCs blocked by B7 antisense peptide.After 3 days or 2 months,T cells were separated from the recipient spleen and the mixed lymphocyte reaction was performed to analyze the response of T cells to the recipient donor-or the third party-pulsed DCs.Meanwhile,3 days or 2 months after immunizing the rats were immunized again by donor-pulsed recipient DCs and the mRNA expressions of cytokines in the spleen were measured by RT-PCR. Results The recipient T cells pretreated with the donor-pulsed recipient DCs blocked by B7 antisense peptide were hyporesponsive to the same donor,but hyperresponsive to the third party via the indirect pathway.Meanwhile,the mRNA expression of IL-10 was increased,but the mRNA expressions of IL-2 and IFN-γ were decreased in the spleen of rats pretreated with the donor-pulsed recipient DCs. Conclusion The DCs blocked by B7 antisense peptide can induce the donor-specific immune hyporesponsiveness by the indirect pathway in vivo,which may be accompolished through the immune deviation.
