摘要
垂体特异性转录因子1((Pituitary Specific Transcription Factor 1,POU1F1)基因是POU基因家族的成员之一,对哺乳动物生长发育、新陈代谢起主要的调控作用。采集116份新疆褐牛血样,利用PCR-RFLP技术检测POU1F1基因第6外显子的多态性,分析不同基因型与生长性状的相关性。结果表明,新疆褐牛的POUIFI基因第6外显子HinfI位点有多态性,新疆褐牛群体在该位点上处于Hardy-Weinberg平衡状态,等位基因A/B的频率分别为0.375/0.625。3种不同基因型间个体出生体质量及1-5月龄体质量的差异不显著(P〉0.05),而BB基因型6月龄体质量与AA基因型差异显著(P〈0.05)。应用Logistic、Gompertz和Von Bertalanffy 3种非线性生长模型对新疆褐牛POU1F1基因3种基因型个体进行生长曲线拟合,所选生长曲线模型均可以较好地预测机体的生长发育情况,拟合优度(R2)均大于0.995。AA、AB、BB基因型3种生长曲线比较,BB基因型具有极限体质量大,生长速率快,拐点体质量及生长拐点月龄大等特点。
POU class 1 homeobox 1(POU1F1) is a member of the tissue-specific POU-containing transcription factor family.Through the PCR-RFLP,the POU1F1 gene with early growth traits were analyzed in 116 Xinjiang Brown Cattle.The results showed that the POU1F1 had three genotypes and two alleles,which were at the Hardy-Weinberg equilibrium.The frequency of alleles A/B in the Xinjiang Brown Cattle population is 0.375/0.625.There was no significant difference in birth weight and 1-5 month weight with the distribution of genotypes(P0.05),but the difference was found in the sixth month weight between AA and BB genotypes(P0.05).The growth curves of AA,AB and BB genotypes were appropriately fitted with Logistic,Gompertz and Von Bertalanffy models.The three models had more effects on fitting with the growth curves,because the indexes of fitness(R2) of the three models were very high(R20.995).Compared the genotypes AA,AB and BB with growth curves,the BB genotypes grew quickly in early growth period,and had the highest weight limitation,the latest growth inflexion.
