过氯酸铵对大鼠体内碘负荷及抗氧化能力的影响 被引量:1

Effects of ammonium perchlorate on iodine load and antioxidant capacity of rats in vivo

摘要 目的探讨过氯酸铵(ammonium perchlorate,AP)对大鼠体内碘负荷和抗氧化能力的影响。方法将SD雄性大鼠24只随机分为对照组、AP低剂量组、中剂量组和高剂量组,染毒剂量分别为130、260、520mg/kg,1次/d,每组6只,染毒13周。于第6周和第13周检测大鼠尿碘水平;并在13周后检测甲状腺组织中总碘量、相对含碘量、甲状腺组织蛋白定量、丙二醛(MDA)、超氧化物歧化酶(SOD)、甲状腺过氧化物酶(TPO)等指标。结果与对照组比较,第6周AP高剂量组尿碘水平明显升高,差异有统计学意义(P<0.05);第13周各剂量组尿碘水平与对照组比较无明显差异;各剂量组甲状腺总碘量和相对含碘量均明显低于对照组(P<0.01);AP中、高剂量组甲状腺总蛋白定量水平明显低于对照组(P<0.05);各剂量组MDA含量和SOD活力与对照组比较均无明显差异;AP高剂量组TPO活力较对照组明显升高(P<0.01)。结论AP可抑制甲状腺摄取碘,在一定时间内造成大鼠尿碘增加,并致甲状腺组织含碘量降低。AP致甲状腺蛋白定量水平减少。AP造成甲状腺过氧化物酶活力升高。 Objective To study the effects of ammonium perchlorate(AP)on iodine load and antioxidant capacity of rats in vivo.Methods Twenty four Sprague Dawley(SD)male rats were randomly divided into three AP treated groups and a control group.Rats in three AP treated groups were exposed to different doses of AP(130,260 and 520 mg/kg/day)through drinking water for 13 weeks.The levels of rat urinary iodine were detected at the 6th and the 13th week.The total iodine in thyroid tissue,relative iodine contents,protein contents in thyroid tissue,contents of malondialdehyde(MDA),and activities of superoxide dismutase(SOD)and thyroid peroxidase(TPO)were measured after the exposure ended.Results Compared with the controls,the level of urinary iodine in high-dose group in the 6th week increased significantly(P0.05);whereas there was no significant change when it was in the 13th week.The total iodine and relative iodine in thyroid tissue of each treated group decreased significantly(P0.01).The total protein levels in high dose group decreased significantly(P0.05).The contents of MDA and SOD activity showed no significant difference.The TPO activity in high-dose group increased significantly(P0.01).Conclusions AP exposure could inhibit thyroid iodine uptake,increase the urinary iodine within a certain period of time and reduce the concentration of iodine in thyroid tissue.AP could induce the compensatory hyperplasia of thyroid tissue and decrease the protein levels of thyroid.AP could also increase the activity of thyroid peroxide.
