摘要
目的研究难治性癫痫大鼠脑组织与外周血p-糖蛋白(P-glycoprptein,pgp)的变化规律并探讨两者的关系。方法 (1)利用苯巴比妥钠(PB)对杏仁核成功点燃的大鼠进行耐药筛选,将其分为药物有效与耐药组大鼠,其后两组动物均每日腹腔内注射PB抗癫痫治疗,同时观察两组大鼠的痫性发作。(2)利用免疫组化及流式细胞仪方法分别于1、2、3、4w测定两组大鼠不同区域脑组织及外周血pgp的表达。结果 (1)除小脑外耐药组大鼠各区域脑组织及外周血pgp的表达显著高于对照组及有效组大鼠(P<0.01,0.05)。(2)除小脑外耐药组大鼠各区域脑组织及外周血pgp表达在1、2、3、4w 4个不同时间点有统计学差异(P<0.01);且两者表达的pgp有随病程增长呈同步增多趋势。结论耐药大鼠外周血pgp水平可作为脑组织pgp表达的简易参考指标。
Objective To explore the variation of P-glycoprptein (pgp)expression in brain tissue and peripheral blood of intractable epilepsy rat and the relationship of them with growth of seizure time. Methods Amygdale-kindled rats screened by PB were divided into drug-effective and drag-resistance groups. Daily seizure attack was observed in two groups while intra-abdominal injection PB was taken. In order to determinate the expression of pgp in the brain and peripheral blood of rats in 1,2,3,4 week respectively, lmmunohistochemistry and flow-cytometry methods were used. Results Pgp expression in peripheral blood and regions of brain tissue except the cerebellum of drug-resistance rats was significantly higher than drug-effective and control ones( P 〈 0.01,0.05 ). Pgp expression in brain regions except cerebellum and peripheral blood of PB-resistant rats showed significant difference in 1,2,3,4 week respectively( P 〈0. 01 ) ,and Pgp expression in brain tissue and peripheral blood had tendency of synchronous rise with course growth. Conclusion Pgp expression in peripheral blood can be used as reference of the expression in brain tissue of drug-resistance rats.
