熟地、鳖甲对大鼠成骨细胞增殖、碱性磷酸酶蛋白和核心结合因子α1 mRNA表达的影响 被引量:2

摘要 目的观察熟地、鳖甲含药血清对大鼠成骨细胞增殖和功能的影响。方法雄性SD大鼠随机分为3组后,分别灌服高浓度熟地、鳖甲煎剂(3.25 g/kg)、低浓度熟地、鳖甲煎剂(1.625 g/kg)及等量容积的蒸馏水,连续灌胃3 d,制得大鼠高、低剂量熟地、鳖甲含药血清及空白对照血清。原代培养并传至第3代经碱性磷酸酶鉴定取得的大鼠成骨细胞,消化计数铺板并分为3组,以上述血清处理72 h,MTT法检测成骨细胞的增殖率,ELISA法检测碱性磷酸酶(ALP)分泌量并以相应OD值进行纠正,并且运用实时荧光定量PCR检测各组成骨细胞核心结合因子α1(Cbfα1)mRNA表达情况。结果低剂量含药血清组成骨细胞增殖率、ALP浓度及Cbfα1 mRNA表达量明显高于其他2组。结论熟地、鳖甲可能是通过提高成骨细胞的增殖潜能而起骨保护作用的。 Objective To observe the influences of medicinal serum of Shudi(Radix Rehmanniae Praeparata) and Biejia(Carapax Trionycis) on the proliferation and function of osteoblasts in rats.Methods SD rats were randomly divided into 3 groups including high-dose medicinal group(high-dose group),low dose medicinal group(low-dose group) and blank group.High-dose group and low-dose group were intragastrically given decoction of Shudi and Biejia(3.250 g/kg and 1.625 g/kg) respectively twice a day and blank group was given equivalent distilled water.The medicinal serum samples of Shudi and Biejia and blank control serum samples were prepared after 3 days.The rat osteoblasts were collected after primary culturing to third generation and identifying with alkaline phosphatase(ALP).All osteoblasts were divided into 3 groups after counting and plating and treated for 72 hours.The proliferation rate of osteoblasts were detected by using MTT test,secretion of ALP was detected by using ELISA and corrected with corresponding OD value,and mRNA expression of core-binding factor α1(Cbfα1)was detected by using RT-PCR in all groups.Results The proliferation rate of osteoblasts,concentration of ALP and expression of Cbfαl mRNA were all higher in low-dose group than those in other two groups.Conclusion Shudi and Biejia can protect bone through promoting the proliferation proficiency of osteoblasts
