摘要
目的建立一种同时测定莪术饮片中3种成分含量的高效液相色谱法。方法固定相为依利特Hypersil ODS柱(4.6mm×250mm,5μm);乙腈-水为流动相,梯度洗脱;流速:1mL/min;柱温25℃;检测波长:216nm;进样量为10μL。结果在上述色谱分离条件下,莪术二酮、莪术醇、吉马酮的浓度在0.008 1~0.257 4、0.006 7~0.213 6、0.001 6~0.050 0mg/mL时,各成分浓度与色谱峰面积之间线性关系良好,平均回收率分别为:99.1%、98.6%、98.2%,回收率RSD分别为:2.3%、1.4%、2.0%。结论本实验研究所建立的方法可靠、灵敏、简便、专属性强,能比较全面反映不同产地莪术饮片的内在质量,为供临床使用的莪术饮片的安全使用及其质量控制提供了科学依据。
OBJECTIVE To establish a high performance liquid chromatography method for the determination of three components in herbal pieces of Rhizoma Curcuma prepared for decoction.METHODS Elite Hypersil ODS column(4.6 mm×250 mm,5 μm) and acetonitrile-water were the stationary and the mobile phases respectively.Flow rate and the column temperature were 1 mL/min and 25 ℃.The detection wavelength was 216 nm and the injection volume was 10 μL.RESULTS The calibration curve for curdione,curcumol and germacrone revealed linearity in the range of 0.008 1~0.257 4,0.006 7~0.213 6,0.001 6~0.050 0 mg/mL,respectively.The average recovery rate was 99.1%,98.6% and 98.2%,with RSD of 2.3%,1.4% and 2.0%.CONCLUSION The method is reliable,simple and accurate,which can be used to identify and evaluate the quality of Rhizoma Curcumae.
