摘要
目的测定延胡索乙素在不同溶剂中的平衡溶解度以及表观油水分配系数,为设计延胡索乙素新剂型提供依据。方法采用HPLC法测定延胡索乙素在6种溶剂中的平衡溶解度以及在正辛醇-缓冲液中的表观油水分配系数。结果延胡索乙素在二氯甲烷及pH 1.2的盐酸溶液中溶解度最高,分别为1 189.88mg/mL及3 304mg/mL;油水分配系数在pH8.0时最高,为394.53。结论延胡索乙素在水中的平衡溶解度及油水分配系数与介质的pH值有关,pH小于2时溶解性较好,当pH>6时,在油相中分配较多。
OBJECTIVE To determine the equilibrium solubility of dl-tetrahydropalmatine(dl-THP) and its partition coefficient for the n-octanol-water/buffer solution system so as to provide a basis for novel exploitation.METHODS A high performance liquid chromatography(HPLC) was established to detect the concentration of dl-THP in water and seven organic solvents and the partition coefficient for the n-octanol-water/buffer solution system.RESULTS The maximum equilibrium solubility was 1 189.88 mg/mL at pH 1.2 and 3 304 mg/mL in dichloromethane,and the maximum of apparent partition coefficient was 394.53 at pH 8.0.CONCLUSION The equilibrium solubility and apparent partition coefficient of dl-THP are correlated to pH of the medium.It dissolves preferably when pH2,and dl-THP distributes more in the lipid phase when pH=8.
