ghrelin抑制ox-LDL诱导的人脐静脉内皮细胞粘附分子表达 被引量:1

摘要 目的探讨不同剂量ghrelin预处理对氧化型低密度脂蛋白(ox-LDL)诱导的人脐静脉内皮细胞粘附分子表达的作用。方法人脐静脉内皮细胞经传代培养后随机分配为对照组、ox-LDL组及1 ng/mL、10 ng/mL与100 ng/mL ghreli预处理组,采用逆转录—聚合酶链反应(RT-PCR)检测各组血管内皮细胞粘附分子-1(VCAM-l)与细胞间粘附分子-1(ICAM-1)的RNA水平变化。结果与空白对照组相比:ox-LDL组内皮细胞VCAM-1与ICAM-1分子的表达水平明显升高(P<0.05)。与ox-LDL组相比,1 ng/mL、10 ng/mL、100 ng/mL ghrelin预处理可使内皮细胞VCAM-1 mRNA含量依次降低33%、52%、63%,内皮细胞ICAM-1 mRNA分子表达水平依次降低18%、46%、57%。结论 ghrelin对ox-LDL诱导的人脐静脉内皮细胞中粘附分子VCAM-1及ICAM-1的表达有明显的抑制作用。 Objective To investigate the inhibitory effect of different concentration of ghrelin on ox-LDL-mediated adhesion molecule expression in human umbilical vein endothelial cells(hUVECs).Methods After subculture,hUVECs were randomly divided into control group,ox-LDL group,ghreli-pretreated group of 1 ng/mL,10 ng/mL and 100ng/mL.RT-PCR were used to detect the RNA changes of vasculareen adhesionmolecule-1(VCAM-l) and inter-cellular adhesion molecule-1(ICAM-1).Results Compared with control group,both VCAM-1 and ICAM-1 expressions were obviously increased(P0.05).While compared with ox-LDL group,ghrelin-pretreated groups showed a decrease in mRNA level of VCAM-1,which were 33%,52% and 63% respectively by different concentrations of 1,10 and 100 ng/mL ghrelin.The mRNA level of ICAM-1 in ghrelin-pretreated groups were also inhibited,with a decrease of 18%,46% and 57% respectively by different concentrations of 1,10 and 100 ng/mL ghrelin.Conclusion Ghrelin could repress the expression of VCAM-1and ICAM-1 induced by ox-LDL in human umbilical vein endothelial cells.
