摘要
目的探讨胆红素对新生儿脐血单核细胞(CBMC)核转录因子-κB(NF-κB)表达的影响。方法选择10例健康足月新生儿作为研究对象,收集脐血,采用明胶/自体血浆贴壁法(gelatin/plasmacoatedflasks)分离CBMC。经含有不同浓度胆红素的牛血清白蛋白溶液孵育1h,再给予脂多糖(LPS)刺激培养细胞,收集单核细胞。采用间接免疫荧光法,观察CBMCNF-κB的活化水平。结果 LPS可促进CBMCNF-κB的活化,而胆红素对其有抑制作用,6mg/dl浓度的胆红素单独即可抑制CBMCNF-κB的活化。先经不同浓度(2.5mg/dl、6mg/dl、9mg/dl、12.9mg/dl、18mg/dl)胆红素孵育,CBMCNF-κB的活化受到不同程度的抑制;再接受LPS刺激,低浓度(2.5mg/d和6mg/dl)胆红素孵育后的CBMCNF-κB的活化可以恢复,高浓度(12.9mg/dl和18mg/dl)胆红素孵育后的CBMCNF-κB的活化仍然受抑制,当胆红素浓度由12.9mg/dl上升至18mg/dl时,CBMCNF-κB的活化受抑制程度的差异,没有统计学意义。随着胆红素浓度的升高,这种抑制作用越明显。结论胆红素可以通过抑制CBMCNF-κB的活性影响单核细胞的免疫功能。
Objective To explore the effects of bilirubin on the expression rate of nuclear factor-kappa B(NF-κB)of cord blood monocyte(CBMC).Methods 10 cases of healthy full-term newborn infants with normal Apgar score were selected randomly as subjects;their monocytes in the umbilical cord blood were in vitro separated by the method of gelatin/plasma coated flasks.The CBMC were preincubated with unconjugated bilirubin dissolved in bovine albumin solution at various levels(0-18 mg/dl).After being preincubated,the CBMC were further cultured with LPS for inducing cellular activation,and then the CBMC were collected.The expression rate of NF-κB was detected by an indirect immuno-fluorescence method.Results The expressive ability of CBMC NF-κB was significantly higher after application of LPS,so LPS caused increase of the expression rate of NF-κB.The bilirubin(6 mg/dl)caused a depression of the expression rate.By comparison,in the presence of LPS,the expression rate of NF-κB of the 2.5 mg/dl preincubated group,preincubated 6 mg/dl group were almost the same as the normal control group(P〉0.05),the expression rate of NF-κB of 12.9 mg/dl preincubated group and 18 mg/dl preincubated group were significantly lower than those of the normal control group and LPS control group(P〈0.05).The expression rate of the 18 mg/dl group was not apparently lower than that of the 12.9 mg/dl group(P〉0.05).The relationship between the bilirubin concentration and the expression rate of bilirubin-treated CBMC was negative correlation,so the inhibition of the expression rate of CBMC will be changed corresponding to the raise of the bilirubin concentration.Conclusions Bilirubin can inhibit the expression rate of NF-κB of CBMC,the mechanism may be involved in immunotoxicity of bilirubin.
