一氧化氮合酶抑制剂对缺血性海马神经元凋亡的抑制作用 被引量:3

摘要 目的 :观察非选择性一氧化氮合酶抑制剂 NG 硝基左旋精氨酸 (L NNA)对缺血性海马神经元凋亡的影响 ,探讨一氧化氮 (NO)在缺血性神经元凋亡中的作用。方法 :2 7只雄性 Wistar大鼠分为假手术组、生理盐水治疗组和 L NNA治疗组。采用 4血管关闭法制成全脑缺血模型 ,利用 DNA10 %聚丙烯酰胺凝胶电泳法和原位末端标记技术 ,观察计数全脑缺血 10分钟再灌注 72小时海马区神经元凋亡情况。结果 :全脑缺血 10分钟再灌注 72小时海马区神经元出现凋亡〔凋亡细胞 (39.0 0± 2 .74)个〕,L NNA可抑制缺血性神经元凋亡〔凋亡细胞 (31.80± 3.71)个 ,P<0 .0 5〕。结论 :NO能诱导缺血性神经元凋亡 ,小剂量反复使用 L Objective:To investigate the effect of N GnitrolLarginine(LNNA) on neuronal apoptosis triggered by global cerebral ischemia in rats.Methods:Global ischemia was induced for 10 minutes in anesthetized rats by quadruple vascular occlusion method.27 male rats were randomly divided into shamoperation control group,salinetreated group and LNNA group.Neuronal apoptosis in hippocampus was examined using DNA electrophoresis and terminal deoxynucleotidyl transferase (TdT)mediated dUTPbiotin nickend labeling(TUNEL assay) 72 hours after reperfusion.Results:Neuronal apoptosis in CA 1 layer of the hippocampus followed global ischemiareperfusion〔apoptosis cell counts apoptosis cell counts(39 00±2 74)cells〕,and LNNA could inhibit neuronal apoptosis 〔apoptosis cell counts (31 80±3 71)cells, P <0 05〕.Conclusions:The results indicate that LNNA is neuroprotective,suggesting that NO may play a role in inducing pyramidal neuron apoptosis in CA 1 layer of the hippocampus as a result of global ischemia.
