星点设计-效应面法优化杉木枝叶中双黄酮的提取工艺 被引量:3

Optimum Extracting Process for Biflavone of the Leaves and Branches of Cunninghamia lanceolata by Central Composite Design and Response Surface Method

摘要 采用星点设计-效应面法优化杉木枝叶中穗花杉双黄酮和金松双黄酮的提取工艺。以乙醇体积分数、料液比、提取时间为自变量,穗花杉双黄酮和金松双黄酮转移率的归一化值为因变量,通过对自变量与因变量的二次多项式拟合,采用效应面法选取较优的工艺条件,并进行预测分析。最终确定穗花杉双黄酮和金松双黄酮的最佳提取工艺为:乙醇体积分数为50%,料液比为1∶13,超声提取3次,每次提取60 min,最佳工艺验证结果与模型预测值相差-2.55%。结果表明:星点设计-效应面法优选的穗花杉双黄酮和金松双黄酮的提取工艺,方法简便、可靠。 The extracting process of leaves and branches of Cunninghamia lanceolata was optimized by central composites design and response surface method.Independent variables were ethanol concentration,solvents ratio and extract time,while dependent variable was the OD value of extraction rates of Amentoflavone and Sciadopitysin.A two-order polynomial equation was fitted to the data,response surface method was used to optimize the extracting process,and the resultant model was used to predict the response in the optimal region.The optimum extraction conditions for Amentoflavone and Sciadopitysin were obtained as follows:the extracting solvent was 50% ethanol in volume,solid-liquid ratio was 1∶ 13,extracting tine was 60 minutes and extracting times was 3.The deviation between observed and predicted values of extraction quantity was-2.55%.The result indicates that the optimum process was simple,convenient and reliable for extracting the leaves and branches of Cunninghamia lanceolata,with a higher precision.
