苯丁酸钠对SHG-44胶质细胞瘤株的抗肿瘤作用机制研究 被引量:2

摘要 目的观察苯丁酸钠对人脑胶质细胞瘤株SHG-44的抗肿瘤作用,探讨其抗肿瘤作用的机制是否涉及调节ASIC2的表达。方法体外培养人脑胶质细胞瘤株SHG-44,平皿克隆形成法测定细胞描定依赖性生长能力,PI染色流式细胞术(Flow cytometry,FCM)分析细胞周期和凋亡率,Hoechst33258染色荧光显微镜观察凋亡细胞的形态,Western Bloting分析药物对ASIC2蛋白表达的影响。结果平皿集落形成法检测显示苯丁酸钠对人脑胶质细胞瘤株SHG-44的抗肿瘤作用,且呈浓度依赖性。PI染色FCM结果表明苯丁酸钠以时间和浓度依赖方式对人脑胶质细胞瘤株SHG-44的抗肿瘤作用。Hoechst33258染色荧光显微镜观察可见核染色质凝集,凋亡细胞呈致密浓染,与对照组相比,苯丁酸钠处理后凋亡细胞比例增加,且呈时间和浓度依赖性。Western Bloting检测显示苯丁酸钠诱导细胞凋亡的机制涉及上调ASIC2表达。结论苯丁酸钠在体外对人脑胶质细胞瘤株SHG-44具有抑制增值的作用,初步推断苯丁酸钠诱发人脑胶质细胞凋亡与其上调ASIC2表达有关。 Objective To observe the anti-tumor effect of benzene sodium butyrate on the brain gliomas strains SHG-44,to discuss whether its antitumor function mechanism is related with adjusting ASIC2 expression.Method The human brain gliomas strains SHG-44 were cultured in vitro and its anchoragedependent growth ability were detected by dish clone forming assay.Flow cytometry(FCM)were employed to analyze the cell cycle and apoptosis,and Hoechst 33258 fluorescence microscope was used to observe the apoptotic cell morphology.Western Bloting method was applied to analyze the effect of drugs on the expression of ASIC2.Results Dish clone forming assay showed that benzene sodium butyrate made a concentration-dependent anti-tumor effect on the brain gliomas strains SHG-44.PI dyeing FCM results show that benzene sodium butyrate effect the glioma SHG strains-44 in a time and concentration-dependent manner.Hoechst33258 dyeing fluorescence microscope could observe the nuclear condensed chromatin.Apoptosis cells were densely and strongly dyed.Compared with the control,the apoptotic cells increased with time and concentration dependence after treated by benzene sodium butyrate.Western Bloting test showed that the mechanism of benzene sodium butyrate inducing apoptosis was related with the up-regulation of ASIC2 expression.Conclusion Benzene sodium butyrate can inhibit the proliferation of human brain gliomas strains SHG-44 in vitro.It was preliminarily inferred that the mechanism of benzene sodium butyrate inducing apoptosis of human brain glial cells which is associated with the ASIC2 expression.
