摘要
目的:探讨金属钴、铬颗粒对人成骨细胞系MG63细胞骨保护素(OPG)及核因子-κB受体活化因子配体(RANKL)表达水平的影响。方法:将金属钴、铬颗粒与人成骨细胞系MG63细胞共培养,ELISA法检测细胞培养上清中OPG、RANKL水平,采用荧光定量PCR法检测OPG、RANKLmRNA表达水平。结果:培养后24、48h实验组RANKL蛋白与mRNA水平显著增加,与对照组相比差异有统计学意义;但OPG蛋白与mRNA水平虽然增加,但与对照组相比差异没有统计学意义;RANKL/OPG比值显著增加,与正常对照组相比差异有统计学意义。结论:金属钴、铬颗粒可以促进RANKL蛋白及RANKL mRNA表达,使RANKL/OPG比率增高,促进破骨细胞分化,从而使骨吸收增强。
Objective: To explore the effectiveness of cobalt,chromium particle on the expression of osteoprotegerin(OPG) and receptor activator of nuclear factor kappa B ligand(RANKL) in human MG63 cells.Methods:MG63 cells were cultured with cobalt,chromium particle.The protein levels of OPG and RANKL were detected by ELISA.The gene expressions of OPG and RANKL in MG63 cells were analyzed by real-time PCR.Results : The protein and mRNA levels of RNAKL in the experimental group were significantly increased.There were significantly different between the experimental group and control group.However,The protein and mRNA levels of OPG were increased,there were no significantly different between the experimental group and control group.The ratio of RANKL/OPG was significantly increased and there were significantly different between the experimental group and control group.Conclusion:Cobalt,chromium particle can promote the protein and mRNA expression of RANKL and then the ratio of RANKL/OPG increases.This may contribute to the differentiation of osteoblasts and the enhancing of bone resorption.
