摘要
目的 探讨强力霉素对碱烧伤后角膜新生血管(CNV)形成及角膜血管内皮生长因子(VEGF)的影响.方法 将35只健康SD大鼠随机选取5只,为A组(正常组).余30只大鼠行右眼角膜碱烧伤,建立角膜新生血管动物模型,将其随机平分为B组(实验对照组)和C组(实验干预组).分别于3d,7d,14d裂隙灯下观察各组CNV的生长情况,计算新生血管面积,并采用免疫组化法检测大鼠角膜组织中VEGF的表达情况.结果 A组没有CNV生成,C组和B组相比CNV的生长明显受到了抑制;免疫组织化学染色结果示:A组VEGF微弱表达,B组和C组的表达明显高于A组,C组VEGF的表达低于B组,且差异具有统计学意义(P〈0.05).结论 强力霉素可有效抑制角膜碱烧伤后CNV的生成及角膜组织中VEGF的表达.
Objective To explore the effects of doxycycline on corneal neovascularization and vascular endothelial growth factor after alkali burn. Methods Five of thirty-five healthy SD rats were randomly selected as group A ( normal group), and the model of corneal neovascularization were established by alkali burn of the right corneas of the other thirty rats which were randomly divided into group B ( experimental control group ) and group C ( experimental intervention group). On the 3rd,7th and 14th day,the growth of CNV were observed while the size of new vessels were fig- ured with the slit lamp, and then the expression of VEGF in the rat corneas was detected in immunohistochemistry meth- od. Results Except group A,both group B and group C showed the growth of CNV,but the growth of CNV in group C was obviously inhibited compared with that of group B ; meanwhile, the result of the immunohistochemistry method showed group A had low expression of VEGF while the expression of VEGF in group B and C was obviously higher than that of group A, but the expression of VEGF in group C was much lower than that in group B, the difference was statistically sig- nificant ( P 〈 0.05 ). Conclusion Doxycycline can effectively inhibit the growth of CNV and the expression of VEGF in the cornea after alkali burn.
