摘要
油茶是我国南方的主要木本食用油料树种,赣南油茶良种是我国重要的油茶资源组成部分。利用SRAP(序列相关扩增多态性)标记技术,对我国油茶良种分子鉴别进行探索性研究。以14个赣南油茶良种为对象,提取叶片基因组DNA作为模板,筛选出稳定性高的引物组合;采用SRAP-PCR的优化反应体系,对获得的清晰可重复的条带进行{0,1}数据转换,并根据SRAP的谱带特征建立了赣南油茶良种的分子鉴别体系,为油茶良种的分子鉴定提供了理论依据和技术基础。
Camellia oleifera is one of main tree species of woody edible oil in South China,and important elite cultivars resource in southern Jiangxi province.The researches on molecular identification of the elite cultivars in south Jiangxi were carried out by SRAP(sequence related amplification polymorphism).By Using the leaf genomic DNA extracted from 14 elite Camellia oleifera cultivars in south Jiangxi province,the improved CTAB method as template,the high-stable primer groups were selected out based on the improved SRAP-PCR system,which can bring specific DNA bands,good polymorphism and high stability.According to the SRAP-based band pattern from these primer groups,the polymorphic DNA bands were transformed in range of {0,1} data,which was used to establish the primary system of molecular identification of elite cultivars from south Jiangxi,thus providing theoretic basis and technique foundation of molecular identification of Chinese elite Camellia oleifera cultivars.
