大鼠视神经挫伤后生长相关蛋白的变化及重组人促红细胞生成素的作用 被引量:1

Changes of growth associated protein and effect of recombinant human erythropoietin in rats after traumatic optic neuropathy

摘要 目的研究大鼠视神经挫伤后视神经生长相关蛋白(GAP-43)的变化及重组人促红细胞生成素(rhEPO)的作用。方法建立外伤性视神经损伤的动物模型,随机分为正常对照组、损伤组(视神经钳夹+生理盐水)及rhEPO组(视神经钳夹+rhEPO),于损伤后1d、4d.7d、14d和28d对各组视网膜及视神经进行形态学观察,免疫组化染色法测定视神经GAP-43的活性。结果rhEPO组较损伤组视网膜及视神经病变明显减轻;在损伤后4d和7d两组间损伤远端GAP-43表达差异无统计学意义(P〉0.05);14d和28drhEPO组损伤远端GAP-43表达较损伤组明显升高(P〈0.05)。结论rhEPO对视神经不完全损伤可促进视网膜神经节细胞存活与视神经纤维再生修复。 Objective To study changes of growth associated protein(GAP-43) and effect of recombinant human erythropoietin (rhEPO) in rats after optic nerves crushed. Method The animal model of traumatic optic neuropathy was established, Sixty-three healthy aduh SD rats without eye diseases were divided into three groups randomly: control group, crush group and rhEPO group. The morphology of retina and optic nerve was observed. Changes of GAP-43 in optic nerve were studied with immunohistochemistry staining at 1d, 4d, 7 d, 14 d and 28 d after crush. Results The pathological change of retina and optic nerve in rhEPO group reduced than in crush group. Immunohistochemistry results showed that there were no significant difference of GAP-43 expression between crush group and rhEPO group ( P 〉 0.05 ) at 4 d, 7 d after crush. However, the expression of GAP-43 in rhEPO group significantly increased than that of crush group at 14 d and 28 d after crush. ( P 〈 0.05 ). Conclusion RhEPO may be helpful in repairing and regenerating of the retina ganglion cells and optic nerve after crush.
