摘要
对 1 9例慢性支气管炎 (慢支 )缓解期患者和1 5例正常对照者经局部支气管肺泡灌洗获取的肺泡巨噬细胞 ( AM) ,加川芎嗪 ( ligustrazine) ,脂多糖( LPS)体外培养 2 4 h后 ,以流式细胞仪分析其CD1 1 c和 CD1 4表达的百分率 ;测定其胞浆游离钙水平 .慢支组 AM膜上 CD1 1 c和 CD1 4的表达均显著高于正常对照组 ;1 0 0 mg· L-1LPS可使慢支组AM膜上 CD1 1 c的表达进一步增加 ;40 0 mg· L-1川芎嗪可明显减少 LPS诱导慢支组 AM膜上CD1 1 c的过度表达 .慢支组 AM胞浆中基础钙水平较正常对照组明显增高 ,但其静息钙水平与正常对照组无显著差异性 ;40 0 mg· L-1川芎嗪明显抑制1 0 mg· L-1LPS诱导慢支 AM胞浆游离钙水平的升高 .结果提示川芎嗪通过抑制慢支患者 AM胞浆游离钙水平升高下调其 CD1 1 c的表达 。
To investigate the effect of ligustrazine on LPS induced CD11c and CD14 expression on alveolar macrophages(AMs) from patients with chronic bronchitis, AMs obtained from 19 patients with stable chronic bronchitis and 15 normal volunteers by bronchoalveolar lavage were purified. The percentages of CD11c and CD14 expression on AMs cultured with LPS, ligustrazine for 24 h in vitro were analysed by flow cytometry and intracellular free calcium concentration([Ca 2+ ] i) in AMs loaded by fura 2/AM was determinated. The results showed that the percentages of CD11c and CD14 expression on AMs from chronic bronchitis group(n=19) were significantly increased compared with normal control group(n=15). High expression of CD11c on AMs induced by LPS(1 mg·L -1 ) from chronic bronchitis group was reduced by adding ligustrazine(400 mg·L -1 ) 10 min before LPS.In contrast with normal control group(n=6), [Ca 2+ ] i in AMs from chronic bronchitis group(n=9) was increased in the absence of extracellular Ca 2+ but not in the presence of 1 mmol·L -1 Ca 2+ ; LPS(10 mg·L -1 ) induced a rapid elevation of [Ca 2+ ] i in AMs that could be completely inhibited by incubating with ligustrazine(400 mg·L -1 ) 10 min before LPS. Ligustrazine reduced expression of CD11c on AMs from chronic bronchitis by inhibiting LPS induced [Ca 2+ ] i elevation in AMs, suggesting that ligustrazine inhibit chronic airway inflammation during stable chronic bronchitis.
