摘要
目的 :采用将集落法和 MTT法结合发展而成的双琼脂 MTT法以克服 MTT法中存在正常细胞干扰测定的缺点。方法 :对人肿瘤细胞及正常细胞株在双琼脂 MTT法和 MTT法光密度、抗癌药抑制率进行比较及临床实体瘤用两种方法的抗癌药抑制率进行比较。结果 :正常细胞在双琼脂 MTT法中生长不良 ,用该两种方法测定临床 2 0例食道癌对抗癌药抑制率 ,比较显示双琼脂 MTT法的抑制率大于 MTT法。结论 :双琼脂 MTT法提高了药敏结果的准确性 ,优于 MTT法。
AIM:To overcome the weakness of interfering by the normal cells in 3 (4,5 dimethylthiazol 2 yl) 2, 5 diphenyltetrazolium bromide (MTT) method, We used double layer agarose MTT (DMTT) method which was developed by combining colony method and MTT method. METHODS: We compared the inhibition rates of the antitumor drugs and the optical density between the human tumor cell lines and the normal cell lines by DMTT method and MTT method, and we also compared the inhibition rates of the antitumor drugs in clinical tumor between the 2 methods. RESULTS: The normal cells grew poorly in DMTT method. It demonstrated that the inhibition rates of the antitumor drugs determined by DMTT method were higher than that of determined by MTT method in 20 esophagus tumors. CONCLUSION: DMTT method improves the accuracy of chemosensitivity and is better than MTT method.
