摘要
纤维二糖酶(cellobiase)是纤维素酶系中的重要组分之一,目前由里氏木霉(Trichoderma reesei)生产的纤维素酶制剂中纤维二糖酶的活力明显偏低,限制了纤维素的糖化效率。本文采用一个黑曲霉菌株(Aspergillus niger ZU-04),在液态发酵条件下生产纤维二糖酶,对主要的发酵工艺参数进行了研究,结果表明,培养基中添加1.0%的麸皮对纤维二糖酶的形成有明显的促进作用;葡萄糖、玉米浆粉的适宜浓度分别为2.0%、0.3%;变温培养缩短了产酶周期,培养4d,酶活力达到最高,为6.23IU/mL。采用黑曲霉纤维二糖酶与里氏木霉纤维素酶协同水解酸预处理后的玉米芯,当纤维素酶用量为20IU/g底物时,纤维二糖酶活力和滤纸酶活力比例为0.43,2d酶解得率达到91.1%。
Cellobiase is one of the important components of cellulase complex.The low amounts of cellobiase in cellulase from Trichoderma reesei limit the rate and efficiency of cellulose saccharification.The main process parameters for cellobiase production by Aspergillus niger ZU-04 under submerged fermentation were investigated.It was found that cellobiase production was increased by supplementing 1.0% wheat bran in the medium.The optimal concentration of glucose and corn steep powder was 2.0% and 0.3%,respectively.The fermentation period was shortened by controlling temperature with the maximum enzymatic activity of 6.23IU/mL after 4d.Cellobiase from A.niger ZU-04 and cellulase from T.reesei catalyzed the hydrolysis of acid-pretreated corncob synergetically.The hydrolysis yield reached 91.1% after 2d when the ratio of cellobiase activity and filter paper activity was 0.43 with cellulase dosage of 20IU/g substrate.
