摘要
目的研究丹参酮Ⅱ-A对大鼠心肌细胞L-型钙通道的影响。方法采用酶解法制备大鼠心肌细胞,应用全细胞膜片钳技术检测大鼠心肌细胞L-型钙通道的电流密度和动力学。结果丹参酮Ⅱ-A对L-型钙通道有抑制作用。在0μmol/L、10μmol/L、20μmol/L和40μmol/L丹参酮Ⅱ-A作下,L-型钙通道的峰值电流密度分别为-13.34±1.06pA/pF、-10.46±0.75pA/pF、-7.62±0.50PA/pF和-4.69±0.18PA/pF,其抑制率分别为28.40%、52.14%和73.50%(n=8,P<0.05)。结论丹参酮Ⅱ-A抑制大鼠心肌细胞L-型钙通道,降低L-型钙通道的电流密度,呈浓度依赖性。
Objective To investigate the effect of TanshinoneⅡ-A on L-type calcium channel in isolated rat ventricular myocytes.Methods rat ventricular myocytes were isolated by enzymatic method,whole cell patch clamp technique was used to record the current and gating.Result Tanshinone II-A inhibit L-type calcium channel current density.Tanshinone Ⅱ-A at 0μmol/L 、10μmol/L、20μmol/L and 40μmol/L,peak L-type calcium channel current density(Ica-L) were-13.34±1.06 pA/pF、-10.46±0.75 pA/pF、-7.62±0.50 pA/pF and-4.69±0.18 pA/pF respectively,the inhibition rates were 28.40%,52.14% and 73.50% respectively(n=8,P0.05).Conclusion Effect of Tanshinone II-A on rat cardiac myocytes L-type calcium current was significantly inhibited.
