摘要
目的探讨SD大鼠静脉血样本在室温下放置时间对血常规测定数据的影响。方法 16只SD大鼠(200~220)g,雌雄各半,采静脉血经EDTA-K2抗凝,置室温(18~25℃),分别于0 min,30 min,1 h,2 h,4 h,8 h,24 h和48 h用MEK-6318K型全自动血液分析仪进行测定。比较不同保存时间对同一静脉血样本血细胞及PLT的影响。结果雌雄SD大鼠血常规数据变化趋势一致,HCT在48 h和0 min比较差异有统计学意义(P<0.05);MCV在24 h、48 h和0 min比较差异有统计学意义(P<0.01);RDW 24 h和0 min比较差异有统计学意义(P<0.05),48 h和0 min比较差异有统计学意义(P<0.01);PLT各时间段和0 min比较差异有统计学意义(P<0.01);PCT30 min、1 h、2 h、4 h和0 min比较差异有统计学意义(P<0.05),其它时间段和0 min比较差异有统计学意义(P<0.01);LYM%48 h和0 min比较差异有统计学意义(P<0.05);MID%24 h、48 h和0 min比较差异有统计学意义(P<0.05);GRN%24 h和0 min比较差异有统计学意义(P<0.05);GRN%48 h和0 min比较差异有统计学意义(P<0.01);其他指标比较差异无统计学意义(P>0.05)。结论 SD大鼠静脉血在常温条件下放置,血常规最好在30 min以后,24 h以内进行测定。
Objective To investigate the effect of storage time on blood routine test of venous blood sample in SD rats at room temperature.Methods Sixteen SD rats(200~220)g were selected,which female and male was half and half,which were adopted Venous blood.Venous blood samples were detected at 0 min,30 min,1 h,2 h,4 h,8 h,24 h and 48 h respectively at room temperature(18~25℃) by using MEK-6318K automatic blood cell analyzer.We compared the effect of different time on blood cells and blood platelet in the same venous blood sample.Results The HCT and LYM% at 48 hours were significantly higher than those at 0 min(P 0.05).The MCV at 24 hours and 48 hours were very significantly higher than those at 0 minute(P0.01).Comparing the RDW and GRN% at 0 minute,the difference was significant(P0.05) at 24 hours and very significant(P0.01) at 48 hours.In contrast with 0 minute,the PLT at different time was significantly higher than the one at 0 minute(P0.01).Comparing the PCT at 0 minute,the difference was significant(P0.05) at 30 min、1 h、2 h and 4 h,very significant(P0.01) at other times.The difference of MID was significant(P0.05) at 24 hours and 48 hours compared to the one at 0 minute.Other hematological parameters in different periods showed no significant differences(P0.05).Conclusion At room temperature,the blood routine test of venous blood in SD rats should be detected within 30 minutes to 24 hours.
