摘要
目的探讨己酮可可碱对人食管癌细胞EC9706的5-Fu化疗增敏作用。方法采用MTT法测定5-Fu及己酮可可碱联合5-Fu对EC9706细胞的抑制作用。结果不同浓度的己酮可可碱联合5-Fu作用于EC9706 12、24、48、72 h,同一时间各组抑制率差异有统计学意义(P均<0.01),同一5-Fu浓度,随着己酮可可碱药物浓度的增加,5-Fu对己酮可可碱细胞的抑制作用也增强(P均<0.01)。25μg/mL 5-Fu联合己酮可可碱(0.5 mg/mL、1.0mg/mL)与50μg/mL 5-Fu对EC9706细胞抑制作用,在24、48、72 h无统计学差异(P均>0.05)。结论己酮可可碱可以增加EC9706细胞对5-Fu的敏感性,减少5-Fu的剂量。
Objective To investigate Pentoxifylline effect on chemotherapy sensitization of 5-Fu in human esophageal carcinoma EC9706 cell. Methods MTT method was used to determine the inhibition effects of 5-Fu and PTX uniting 5-Fu to EC9706 Cell line. Results The EC9706 cells were treated by different concentration of 5-Fu and 5-Fu uniting PTX for 12 h, 24 h, 48 h and 72 h. There were significant difference in inhibition rates in each group at the same time(P 〈0.01 ). The inhibition of 5-Fu to EC9706 cells was gradually increasing along with the ascending of PTX concentration ; There was no significant difference between the 0.5 mg/mL PTX or 1.0 mg/mL PTX uniting 5-Fu 25 μg/mL and only 50 μg/mL 5- Fu at 24 h, 48 h and 72 h ( all P 〉 0.05 ). Conclusion PTX makes the EC9706 cells more sensitive to 5-Fu, and decrease dose of 5-Fu. e
