摘要
目的建立以高效液相色谱法测定人血浆中奥沙西泮的浓度。方法血浆样品用乙醚萃取,在55℃水浴中用氮气吹干,残留物用甲醇溶解后进样。色谱柱为Shim-pack CLC-CN(150 mm×6.0 mm,5μm),流动相为正己烷-甲醇-无水乙醇(80:17:3,V/V/V),流速为1.10 mL·min-1,柱温为30℃,紫外检测波长为230 nm。结果奥沙西泮在0.02~5.0 mg.L-1范围内线形关系良好Y=7.163×10-3+1.030C(r=0.999 6)。方法回收率近100%,日内和日间RSD均小于5%(n=5)。结论本方法适用于血浆中奥沙西泮的检测。
Objective To establish a method for determination of the concentration of oxazepam in human plasma by HPLC. Methods The plasma samples were extracted with ether. The ether extraction was evaporated under nitrogen stream at 55℃ in water bath, and the residue was dissolved in methanol. Shim-pack CLC-CN (150 mm×6.0 mm, 5txm) was used as the analytical column. Mobile phase consists of n-hexane:methanol: ethanol(80:17:3, V/V/V), with a flow rate of 1.10 mL.min-1. The temperature of chromatographic separation was 30℃, and the drug was detected at 230 nm by an ultraviolet detector. Results A good linearity was obtained from 0.02 to 5.0 mg'L-1 with a correlation of 0.9996. The recovery of oxazepam was nearly 100%. Intraday RSD and interday RSD were both less than 5% (n=5). Conclusion The method is suitable for monitoring oxazepam in human plasma.
