摘要
对红酵母 RY-98菌株产生类胡萝卜素的培养基组成和培养条件进行了初步研究 ,并对发酵过程作了动态分析 .结果表明 :培养基组成、糖浓度和添加剂以及培养基初始 p H值、摇瓶装量与培养时间等 ,均对该菌细胞生物量和类胡萝卜素产量有影响 ,其中 ZS添加剂的加入可以明显促进菌体类胡萝卜素的形成 .在初步优化的培养基组成 (葡萄糖 4 0 g/L,玉米浆 1 5g/L,( NH4) 2 SO42 g/L,ZS添加剂 1 g/L)和培养条件 (培养基初始 p H6.0 ,装量 60 m L/2 50m L摇瓶 ,2 8℃培养 60 h)下 ,菌体生物量及类胡萝卜素含量分别为 2 7mg/m L和 4 98.2 μg/g.通过观测发酵过程生理现象的变化 ,确认该菌类胡萝卜素是菌体的次生代谢产物 ,主要形成于对数生长期的末期和稳定期 .
The fermentation medium of carotenoids from RY 98 strain of Rhodotorula was selected. The compositions of optimal medium were as follows: glucose 40 g/L, corn steep liquor(CSL)15 g/L, (NH 4) 2SO 4 2 g/L, additive ZS 1 g/L, and initial pH6 0. The carotenoids of RY 98 strain showed a distinct increase if ZS added in the medium. The optimal medium volume in 250 mL flask was 60 mL. The dry cells of 27 mg/mL and the carotenoids of 498 2 μg/g were produced by strain RY 98 with above mentioned conditions after growing at 28 ℃ for 60 h. By examining the growth pH,the synthesis of carotenoids and carbohydrate source using, considering that the carotenoids is coming only from the later stage of the log growth phase and the stationary phase of strain RY 98.
