摘要
目的:观察五味子乙素(Sch-B)对二氧化硅致大鼠矽肺中基质金属蛋白酶2(matrix metallopro-teinase,MMP-2)和其抑制物基质金属蛋白酶组织抑制因子1(tissue inhibitor of matrix metalloproteinase,TIMP-1)含量的影响。方法:96只大鼠随机分为对照组、染尘组和Sch-B干预组,每组32只,采用暴露式气管内注入SiO2混悬液法建立大鼠矽肺模型,造模后1 d灌胃给予Sch-B干预治疗,分别在药物干预3,7,14,28 d时处死大鼠,取其肺组织,HE染色检测肺组织病理改变;ELISA法测定大鼠肺组织中MMP-2和TIMP-1蛋白的含量。结果:与对照组相比,染尘组MMP-2和TIMP-1蛋白含量在3,7,14,28 d时明显增高(P<0.01);与染尘组相比,Sch-B干预组MMP-2和TIMP-1蛋白含量均明显降低(P<0.01),但仍显著高于对照组(P<0.01),差异有统计学意义。结论:Sch-B对二氧化硅致大鼠矽肺中不同时间点肺组织MMP-2和TIMP-1的表达均有一定程度的抑制作用,能在一定程度上调整MMP-2和TIMP-1的比值使其趋于平衡,具有减轻二氧化硅致矽肺大鼠肺纤维化的作用。
Objective:To investigate the effect of schisandrin B(Sch-B) on the contents of matrix metalloproteinase 2(MMP-2) and tissue inhibitor of matrix metalloproteinase(TIMP-1) protein in lungs of rats silica-induced silicosis.Methods:Ninty-six Wistar rats were randomly divided into control group,silica group and Sch-B group,with 32 rats in each group.The rat silicosis model was established by a single intratracheal injection of silica.From the second day of model establishment,rats in Sch-B group were treated with Sch-B for 3,7,14 and 28 days respectively,then the rats were sacrificed and lungs were collected.Histopathological changes of lungs were observed by HE staining.The contents of MMP-2 and TIMP-1 protein were determined by ELISA.Results:Compared with control group,the contents of MMP-2 and TIMP-1 of silica group were significantly higher on d3,d7,d14 and d28(P0.01).Compared with silica group,the contents of MMP-2 and TIMP-1 of Sch-B group were significantly lower(P0.01),but higher than control group(P0.01).Conclusion:Sch-B can inhibit the expressions of MMP-2 and TIMP-1 protein in rats with silica-induced silicosis at every time point.Sch-B can adjust MMP-2/TIMP-1 ratio and reduce silica-induced pulmonary fibrosis in rats.
