氟对大鼠脑Cu/Zn-SOD蛋白表达的影响及硒的拮抗作用 被引量:1

摘要 目的通过动物实验,从分子水平观察Cu/Zn-SOD蛋白表达与氟中毒的关系及硒对氟的拮抗作用。方法 48只3周龄SD大鼠,雌雄各半,按体重随机分为6个组。对照组每天给予去离子水灌胃,氟暴露组每天给予氟化钠10、20和40 mg/kg灌胃,染氟3个月。加硒拮抗组氟化钠(20 mg/kg)灌胃同时自由饮用加入亚硒酸钠(2 mg/L)的去离子水,加硒组自由饮用去离子水加入亚硒酸钠(2 mg/L)。染毒3个月后处死。测定大鼠脑组织超氧化物歧化酶(SOD)活性,采用免疫组化法检测脑组织SOD表达(IOD值)。结果①与对照组比较,氟暴露各组大鼠脑组织超氧化物歧化酶(SOD)活性逐渐降低(P<0.05),加硒拮抗组较氟暴露组明显升高(P<0.05),加硒组明显高于各组;②氟暴露组脑组织Cu/Zn-SOD蛋白表达逐渐减少(P<0.05),加硒拮抗组较氟暴露组明显升高(P<0.05),加硒组明显高于各组。结论氟可通过降低Cu/Zn-SOD蛋白引起机体脂质过氧化损伤,硒可拮抗氟引起的氧化损伤。
