优化溃结方对溃疡性结肠炎大鼠白细胞介素-1β的影响 被引量:5

Effects of optimized ulcerative colitis decoction on IL-1β content and protein expression in ulcerative colitis rats

摘要 目的研究优化溃结方对溃疡性结肠炎大鼠模型结肠组织白细胞介素1β(IL-1β)含量及蛋白表达的影响。方法健康SD大鼠50只,随机分为正常组、模型组、补脾益肠丸组、柳氮磺吡啶组、优化溃结方组。TNBS/乙醇法建立溃疡性结肠炎大鼠模型。从造模d 4开始,正常组和模型组用生理盐水3 mL灌胃,每日1次;补脾益肠丸组按1.62 g.kg-1剂量,给予浓度108 g.L-1补脾益肠丸3 mL灌胃,每日1次;柳氮磺吡啶组按0.36 g.kg-1剂量,给予浓度24 g.L-1柳氮磺吡啶3 mL灌胃,每日1次;优化溃结方组按8.37 g.kg-1剂量,给予浓度558 g.L-1优化溃结方3 mL灌胃,每日1次,均连续灌胃21 d。各组灌胃给药后,检测溃疡性结肠炎模型大鼠结肠组织IL-1β含量和IL-1β蛋白表达。结果模型组与正常组比较,IL-1β含量升高、蛋白表达升高,差异有显著意义(P<0.01);补脾益肠丸组、柳氮磺吡啶组、优化溃结方组与模型组比较IL-1β含量、蛋白表达均明显下降,差异有显著意义(P<0.01);优化溃结方组与补脾益肠丸组比较,IL-1β含量、蛋白表达均明显下降,差异有显著意义(P<0.01);优化溃结方组与柳氮磺吡啶组比较,IL-1β含量、蛋白表达均无显著差异(P>0.05)。结论优化溃结方可以降低溃疡性结肠炎大鼠模型中结肠组织IL-1β含量和蛋白表达。 AIM To study the effects of optimized UC decoction (OUCD) on IL-1β content and protein expression in ulcerative colitis (UC) rats. METHODS A total of 50 healthy SD rats were randomly divided into normal group, model group, Bupiyichang pill group, sulfasalazine (SASP) group and OUCD group. From the fourth day after the establishment of ulcerative colitis model by TNBS/ethanol method, rats in normal group and model group were administered intragastrically with normal saline 3 mL once daily for 21 days, rats in Bupiyichang pill group, SASP group and OUCD group were administered intragastrically with 108 g·L^-1 Bupiyichang pill 3 mL (1.62 g·kg^-1), 24 g·L^-1SASP 3 mL (0.36 g·L^-1) and 558 g·L^-1 OUCD 3 mL (8.37 g·kg^-1) respectively, for 21 days. IL-1β content and protein expression in colon tissue were examined after intragastric administration. RESULTS Compared with the normal group, IL-1β content and protein expression of the model group rats increased significantly (P 〈 0.01). Compared with the model group, IL-1β content and protein expression of the Bupiyichang pill group, SASP group and OUCD group decreased significantly (P 〈 0.01). IL-1β content and protein expression in the OUCD group were lower than those in Bupiyichang pill group (P 〈 0.01 ). There was no significant difference of IL-1β content and protein expression between the OUCD group and SASP group (P 〉 0.05). CONCLUSION OUCD could reduce the IL-1β content and protein expression in colon tissue in ulcerative colitis rats.
