摘要
目的β-细辛醚对核转录因子κB(nuclear factor Kappa B,NF-κB)表达的影响,为进一步应用开发打下基础。方法培养RAW264.7细胞,脂质体LipofectamineTM2000转染NF-κB启动子双荧光素酶报告基因分析系统Luc2P/NF-κB-RE/Hygro,0.2μg/mL LPS刺激,以15、150μmolβ-细辛醚干预,孵育24 h后检测相对荧光素酶活性;按以上分组干预培养RAW264.7细胞,采用蛋白免疫印迹法检测NF-κB蛋白表达。结果荧光素酶报告基因结果表明,β-细辛醚有抑制NF-κB启动子活性作用,且随剂量增大抑制作用增强;蛋白免疫印迹法结果表明,β-细辛醚有抑制NF-κB蛋白表达作用,且随剂量增大抑制作用增强。结论β-细辛醚能抑制NF-κB转录与表达,可能有较强的抗炎作用。
Objective To investigate the effect of β-asarone on nuclear factor Kappa B(NF-κB).Methods RAW264.7 were cultured and transfected with LipofectamineTM 2000 and the pGL4.32(luc2P/NF-κB-RE/Hygro) Vector as the protocol.24 h after treatment with 0.2 μg/mL LPS and 15/150 μmol/L β-asarone,relative activity of NF-κB Luciferase report gene was detected,and the expression of NF-κB p65 was assayed with Western blot method.Results The results of relative activity of NF-κB Luciferase report gene conformity with that of Western blot,showed that β-asarone inhibited relative activity of NF-κB Luciferase promoter report gene and NF-κB p65 expression in RAW264.7 induced by LPS.Conclusion β-asarone could inhibit both transcription and translation of NF-κB p65.
