摘要
目的:探讨FTY720抑制人乳腺癌MCF-7细胞和喉癌Hep-2细胞增殖作用。方法:将人乳腺癌MCF-7细胞和喉癌Hep-2细胞用不同浓度FTY720作用48 h,MTT法检测细胞活性,流式细胞术检测其对细胞周期及凋亡率的影响。结果:MTT结果显示,FTY720对乳腺癌MCF-7细胞和喉癌Hep-2细胞增殖均有抑制作用,而且对喉癌Hep-2细胞抑制作用的浓度依赖性高于对乳腺癌细胞的作用;流式细胞术检测结果显示,FTY720可将乳腺癌MCF-7细胞阻滞于G1期,将喉癌Hep-2细胞阻滞于G2/M期,并明显促进乳腺癌MCF-7细胞凋亡。结论:一定浓度的FTY720能明显抑制体外培养的乳腺癌和喉癌细胞增殖,调节其细胞周期,诱导乳腺癌MCF-7细胞凋亡。
Objective: To explore the inhibiting effect of FTY720 on proliferations of human breast cancer MCF - 7 ceils and laryn-geal carcinoma Hep -2 cells. Methods: Human breast cancer MCF -7 cells and laryngeal carcinoma Hep -2 cells were treated with different concentrations of FTY720 for 48 hours, MTr method was used to detect cellular activity, flow cytometry was used to detect the impact on cell cycle and apoptosis rate. Results: The .results of MTT method showed that FTY720 could inhibit proliferations of human breast cancer MCF - 7 ceils and laryngeal carcinoma Hep - 2 ceils, and the concentration - dependence of inhibiting effect on laryngeal carcinoma Hep - 2 cells was stronger than that on breast cancer MCF - 7 cells. The results of flow cytometry showed that FTY720. could arrest breast cancer MCF -7 cells at G1 phase and arrest laryngeal carcinoma Hep -2 ceils at G2/M phase, and FTY720 could promote apoptosis of breast cancer MCF - 7 cells. Conclusion: A certain concentration of FTY720 can inhibit proliferations of human breast cancer cells and laryngeal carcinoma cells in vitro, regulate cell cycle, and induce apoptosis of breast cancer MCF - 7 cells.
