摘要
以羧甲基纤维素钠为惟一碳源,从土壤中筛选出一株产纤维素酶的菌株XW-13,其摇瓶培养液的CMC酶活力为15.05μg/mL,滤纸酶活力为15.13μg/mL。酶学性质试验表明,该酶的最适反应pH 7.0,在pH 5.0~9.0时有较好的稳定性,酶活力保持60%以上。该酶的最适反应温度为40℃,在温度为20℃时基本稳定,保温2 h仍有80%以上的活力。在化合物浓度为0.2 mg/mL的条件下,NH4+、Na+、Fe2+、K+、Ca2+、Mn2+、Zn2+、Mg2+和Li+对酶活力有增进作用,Hg+和Cu2+对酶活力有抑制作用。
A cellulase producing strain XW-13 was isolated from soil.The CMC enzyme activity was 15.05 μg/mL,and the filter paper activity reached 15.13 μg/mL.Then the physiological and biochemical characteristic of the enzyme were determined by relevant experiments.Enzymatic properties experiments showed that the optimum pH for the cellulase was 7.0 and its activity was above 60% when pH was from 5.0 to 9.0.The optimum reaction temperature was 40 ℃.It kept stable when the temperature was below 20 ℃,NH4+,Cu2+,Na+,Fe2+,K+,Ca2+,Mn2+,Zn2+,Mg2+,and Li+ could promote the activity of the enzyme,while Cu2+ and Hg2+ could restrain the activity.
