摘要
目的研究下调CD147表达后宫颈癌细胞系HeLa细胞中survivin表达及凋亡的变化。方法设计、合成两对CD147编码基因的反向重复序列,运用瞬间转染方法抑制HeLa中CD147表达,应用RT-PCR、Western blotting方法检测干扰后CD147、survivin、Bim及caspase-3表达改变,流式细胞术检测干扰后肿瘤细胞的凋亡情况。结果 SiR-NA sequence 1、2均有效抑制CD147基因表达(P<0.05),干扰后的survivin表达明显下降,干扰后caspase-3mRNA水平升高(P<0.05),活性caspase-3蛋白及Bim水平增高(P<0.05),肿瘤细胞凋亡增加,以细胞早期凋亡改变最为明显(P<0.05)。结论沉默CD147基因表达可下调HeLa细胞中survivin的表达,诱导肿瘤细胞凋亡。
Objective Inhibition of CD147 expression would affect HeLa cell apoptosis.Methods The method of RNAi was used to down-regulated CD147 expression in HeLa cells,and then the technologies of RT-PCR and western blotting were used to detect expression of CD147,survivin,Bim and caspase-3,the percentage of cell apoptosis were detected by FACS.Results SiRNA sequence 1,2 inhibit the CD147 expression in HeLa effectively(P〈0.05),and followed the down-expression of Survivin(P〈0.05),the up-expression of caspase-3 and Bim(P〈0.05);the percentage of tumor cells' apoptosis increased,and the early apoptosis was the most obvious(P〈0.05).Conclusion Inhibition of CD147 expression induced the down-expression of Survivin,and induced the tumor cell apoptosis.
