摘要
目的 :探讨氟的生殖遗传毒性。方法 :应用单细胞凝胶电泳试验 (SCGE)研究了氟化钠对离体小鼠睾丸细胞的DNA损伤与修复效应。结果 :氟化钠在 (0 .6 2~ 5 ) mm ol/ L 之间可引起小鼠睾丸细胞 DNA单链断裂 ,出现拖尾的彗星细胞。无论是拖尾细胞百分率 ,还是 DNA迁移长度都随氟化钠浓度的增加而增加 ,表现出明显的计量反应关系。此外 ,氟化钠所致的睾丸细胞 DNA损伤基本在 2 h内完全修复。结论 :在一定浓度下 ,氟化钠可引起离体小鼠睾丸细胞的 DNA损伤 ,可能是一种潜在的雄性生殖毒物。
Objective:To study the genotoxicity of fluoride.Method:The effects of sodium fluoride on DNA damage and repair in isolated male mice testicular cells were observed with single cell gel electrophoresis test(Comet assay).Results:At the concentriation of 0 62mmol/L to 5mmol/L,sodium fluoride could induce the breakage of DNA single strand and resulted in comet cells with tail.Both the number of comet cells and the length of DNA migration increased with the increasing of concentration of sodium fluoride,showing the obvious dose effect relationship.In addition,the DNA damage could be repaired basically within two hours incubation.Conclusion:The research suggested that at certain concentrations,sodium fluoride could induce DNA damage in isolated male mice testicular cells and might be a kind of male reproductive toxicants.
