人脐带间充质干细胞对阿尔茨海默病模型小鼠学习记忆能力的影响 被引量:4

摘要 目的观察人脐带间充质干细胞(human umbilical cord mesenchymal stem cells,hUC-MSCs)对化学损伤(D-半乳糖+氯化铝)及海马物理损伤的拟阿尔茨海默病(Alzheimer's disease,AD)模型小鼠学习记忆能力的影响。方法 90只昆明小鼠采用D-半乳糖120 mg/(kg.d)、氯化铝20 mg/(kg.d)共同诱导10周以及海马物理损伤的方法分别复制出2种不同的拟AD小鼠模型。实验分为5组:化学损伤模型对照组、物理损伤模型对照组、化学损伤+hUC-MSCs治疗组、物理损伤+hUC-MSCs治疗组,另设一个空白对照组。hUC-MSCs移植治疗一段时间后,用Morris水迷宫和跳台仪分别检测小鼠学习记忆能力。结果与化学损伤模型对照组比较,化学损伤+hUC-MSCs治疗组小鼠水迷宫逃避潜伏期显著缩短(P<0.05),跨越平台次数显著增加(P<0.05);跳台错误次数显著减少(P<0.05),潜伏期显著延长(P<0.05)。但物理损伤+hUC-MSCs治疗组与对应的模型对照组比较差异无统计学意义(P>0.05)。结论侧脑室注射hUC-MSCs对化学性损伤拟AD小鼠学习记忆能力有改善作用。 Objective To investigate the effects of human umbilical cord mesenchymal stem cells (hUC - MSCs) on the learning ability and memory of mice with Alzheimer's disease (AD), which was induced by D -galactose (D - gal) and aluminium chloride (A1C13 ) , or hippocampus physical injury. Methods Ninety Kunming mice were randomly divided into five groups : blank control ( Group A) , chemical damage model control ( Group B ) , physical injury model control ( Group C) , chemical damage + hUC - MSCs treatment ( Group D) , and physical injury + hUC - MSCs treat- ment groups ( Group E). Intraperitoneal injection of D - gal [ 120 mg/( kg ~ d) ] and oral administration of nlC13 [ 20 mg/ (kg ~ d) ] were given for conduction of chemical damage model for 10 weeks; while physical hippoeampus injury was in- duced using curved syringe needle. Morris water maze and jumping apparatus were used to test the ability of learning and memory of the subjects. Results Signifienat improvement in Morris water maze test, including shorter escape latency and more platform - crossing frequencies, and in jumping apparatus, including less jumping stand error number ( EN ) and longer step - down latency (SDL), were observed in Group D when comparing with those in Group B (P 〈0. 05). No sig- nificant difference was revealed between Group E and Group C ( P 〉 0.05 ). Conclusion Lateral ventricle injection of hUC - MSCs could improve the learning ability and memory of chemical damage - induced AD - like mice.
