摘要
采用阴离子交换层析法,对γ-PGA进行分离纯化。通过阴离子树脂的静态吸附、动态实验,得出了上样的最佳pH8.0,上样量27.3mg/g树脂,洗脱液为0.7mol/L NaCl溶液,洗脱流速1.0mL/min,洗脱温度20℃。最终γ-PGA的纯度由86.7%提高到96.2%。
y-PGA was purified by anion exchange column chromatography. Through statical adsorption test and dynamic test of the anion resin,the optimum technological conditions were determined as follows.sample pH 8.0,sample quantity of resin 27.3mg/g ,eluant 0.7 mol/L NaCI,eluting velocity 1.0mL/min,and eluting temperature 20℃. At last,the purity of y-PGA was improved from 86.7% to 96.2%.
