小半夏加茯苓汤不同提取部位总生物碱含量研究

摘要 目的研究小半夏加茯苓汤不同提取部位总生物碱的含量差异,为制定小半夏加茯苓汤的提取工艺和质量控制标准奠定基础。方法采用紫外分光光度法测定小半夏加茯苓汤醇提物不同极性溶剂萃取部位总生物碱的含量,以溴百里酚蓝作为显色剂,最大吸收波长为410 nm。结果小半夏加茯苓汤不同提取部位的总生物碱在0.012 2~0.122 0 mg/ml(r=0.999 7)范围内呈良好线性关系,其正丁醇萃取部位总生物碱含量占23.254 0%、水层总生物碱含量占10.744 8%、石油醚层总生物碱含量占7.259 4%、乙酸乙酯层总生物碱含量占8.201 9%。结论小半夏加茯苓汤不同提取部位总生物碱含量差异较大,以正丁醇萃取部位含量最高,其次是水层、石油醚层、乙酸乙酯层,即各部位总生物碱含量由高至低依次为正丁醇萃取部位、水层、石油醚层、乙酸乙酯层,为进一步研究小半夏加茯苓汤的止呕药效部位奠定了基础。 Objective To study the difference of the total alkaloid content among the different extractive fractions in Xiaobanxia-Fuling Decoction to lay the foundation for fomulating its extraction technology and quality control standards.Methods The ultraviolet spectrophotometry method was used to measure the total alkaloid content in ethanol extract of Xiaobanxia-Fuling Decoction with different polar solvent extraction,with bromothymol blue as color reagent and the maximum absorption wavelength of 410nm.Results The total alkaloid contents of different extractive fractions in Xiaobanxia-Fuling Decoction showed good linear relation in the range of 0.012 2-0.122 0mg/ml(r=0.999 7),the total alkaloid content was 23.254 0% in the n-butanol fraction,10.744 8% in the water layer,7.259 4% in the petroleum ether layer and 8.201 9% in the ethyl acetate layer.Conclusion The total alkaloid contents of the different extracted fractions in Xiaobanxia-Fuling Decoction are quite different,especially the n-butanol extraction fraction has the highest contents,followed by in turn the water layer,petroleum ether layer and ethyl acetate layer,that is,n-butanol extracted fraction water layerpetroleum ether layer ethyl acetate layer,which lays the fundation for further study of the pharmaceutical part stopping vomiting in Xiaobanxia-Fuling Decoction.
