PCNA和P21蛋白在子宫内膜癌中的表达及意义 被引量:9

Expression and significance of proliferating cell nuclear antigen and P21 protein in endometrial cancer

摘要 目的探讨P21蛋白和增殖细胞核抗原(proliferating cell nuclearantigen,PCNA)在子宫内膜癌中的表达及意义。方法应用免疫组织化学SP法检测正常子宫内膜组织20例(对照组)、子宫内膜非典型增生组织9例(非典型增生组)和子宫内膜腺癌组织57例(子宫内膜癌组)中P21蛋白和PCNA的表达情况。结果对照组P21蛋白阳性表达率明显高于非典型增生组与子宫内膜癌组(P<0.05),子宫内膜癌组PCNA高增殖率明显高于对照组与非典型增生组(P<0.05);P21蛋白阳性表达率及PCNA高增殖率在子宫内膜组织的手术-病理分期、分化程度及淋巴结转移方面差异有统计学意义(P<0.05);子宫内膜癌组PCNA高增殖率P21表达阳性者为37%,P21表达阴性者为80%,差异有统计学意义(P<0.05);子宫内膜癌组织P21蛋白阳性表达率与PCNA高增殖率呈负相关(r=-0.414,P=0.002)。结论 P21蛋白和PCNA异常表达可能在子宫内膜癌的发生、发展中起重要作用。 Objective To explore the expression and significance of P21 and proliferating cell nuclear antigen(PCNA) protein in endometrial cancer.Methods Immunohistochemical SP method was used to detect the expressions of P21 and PCNA protein in 20 cases of normal endometrium(control group),9 cases of endometrial atypical hyperplasia(atypical hyperplasia group) and 57 cases of endometrial carcinoma(endometrial cancer group).Results The positive rate of P21 protein was obviously higher in control group than that in the other groups(P〈0.05).The high proliferation rate of PCNA protein was higher in endometrial cancer group than that in the other groups(P〈0.05).The positive rate of P21 protein and the high proliferation rate of PCNA were significantly different in operation stage,pathology stage,differentiation degree and lymph metastasis(P〈0.05).The high proliferation rate of PCNA was 37% in positive P21 protein and 80% in negative P21 protein in endometrial carcinoma group,which showed a significant difference between them(P〈0.05).The positive rate of P21 was negatively correlated with the high proliferation rate of PCNA(r=-0.414,P=0.002).Conclusion The abnormal expressions of P21 and PCNA protein may play an important role in carcinogenesis and development of endometrial cancer.
