摘要
目的观察丹参对肝癌大鼠模型缺氧诱导因子-1α(HIF-1α)、微血管密度(MVD)的影响。方法以二乙基亚硝胺(DEN)诱导肝癌大鼠模型,用酶联免疫法检测血清HIF-1α的含量,免疫组织化学法检测大鼠肝癌组织HIF-1α的表达,用CD34标记免疫组织化学法检测微血管密度。结果与模型组比较,丹参中、高剂量组能明显降低血清HIF-1α的浓度;免疫组织化学法结果显示,随着丹参剂量的增加,肝癌组织HIF-1α的表达逐渐减弱,MVD逐渐降低。结论丹参可以抑制HIF-1α的表达,降低MVD,进而抑制肝癌细胞的生长和转移。
Objective To investigate the effect of Salvia miltiorrhiza on the expression of hypoxia inducible factor-1a(HIF-1a) and microvessel density (MVD) in rat models of hepatocellular carcinoma. Methods Rat models of hepatocellular carcinoma were induced by diethylnitrosamine. The level of serum HIF-1a was detected by enzyme linked immunosorbent assay. The expression of HIF-1a and the MVD were detected by immunohistochemical assay. Results Compared with model group, the level of serum HIF-let was decreased in Salvia miltion'hiza middle-dose group and Salvia mihiorrhiza high-dose group respectively, and the difference was significant. The immunohistochemical result showed that the expression of HIF-1a and the MVD gradually decreased with the increase of Salvia miltiorrhiza dose. Conclusion Salvia miltiorrhiza could inhibit the growth and metastasis of hepatoma carcinoma cell by weakening the expression of HIF-1a and decreasing MVD.
