摘要
目的 探索苯丙氨酸预防自发性高血压大鼠(SHR)病理性心血管肥厚的机理。方法 将断奶雄性SHR分为实验组和对照组各10只。前者给予含3%L-苯丙氨酸的标准饲料,后者给予标准饲料,在标准条件下喂养8周。观察L-苯丙氨酸对SHR心血管组织病理变化的影响。 结果 L-苯丙氨酸可预防SHR的血压升高,使体重增加,心脏重量减轻,心/体指数明显下降。镜检显示:实验组SHR主动脉管壁中层厚度变薄,血管及心室肌组织胶原含量少,弹力板较薄,心肌细胞浆内肌原纤维排列整齐,线粒体结构完整。结论 苯丙氨酸对SHR心血管组织有保护作用,可预防SHR血压升高及其心血管组织病变的发生。
Objective To explore the mechanism of preventive effects of phenylalanine (Phe) on cardiovascular hypertrophy in spontaneously hypertensive rats (SHR). Methods 20 weanling male SHRs were randomly divided into two groups. The experimentals were fed with chow supplemented with 3% Phe, the controls with standard -- chow for 8 weeks in standard conditions. It was performed to measure the blood pressure of the rats, observed the pathological changes of cardiac and vascular tissues with light and trans -- eclectron-- microscopy, analysed the collagen content of the myocardium with digital video -- frequency processing technique. Results Phe. could prevent blood pressure to rise with age, body weight increase, heart weight decrease and thinning the aortic media. The collagen content in the myocardium of the control was also decreased significantly. Under light microscope, the structure of the intima and adventitia of the vessel wall were intact, collagen and elastin contents were decreased, and myocardium injury was also improved. Under trans -- electronic -- microscope, there were less myofibrillae and few collagen fibrillae in VSMC. The elastic lamella in the intercellular space became thinner and the mitochondria stucture was intact. Cardiac myofibrillae appeared normal. The myotome and mitochondria remained in their normal appearance. Conclusion Phenylalanine benefit on the cardio -- vasculr tissue prevents pathological changes in SHR.
