摘要
目的:比较下瘀血汤全药与组分处方对免疫性肝纤维化大鼠肝组织I型胶原α1多肽链(COL-Iα1)、基质金属蛋白酶抑制因子1(TIMP-1)表达的作用差异,探讨下瘀血汤全药与组分处方抗肝纤维化的作用机制。方法:Wistar大鼠随机分为正常组、造模组。造模组大鼠腹腔注射猪血清复制肝纤维化模型,每次0.5 ml/只,每周2次,共12周。造模成功后造模组大鼠随机分为模型组、下瘀血汤组、组分处方组,第8周开始下瘀血汤组、组分处方组大鼠造模同时灌胃给药,按每千克体质量10 ml的容积,下瘀血汤组每日2.34 g/kg,组分处方组每日0.648 g/kg,连续4周。正常组、模型组大鼠给予同体积0.9%NaCl溶液。12周末,处死动物,获取标本,实时荧光定量PCR检测肝组织COL-Iα1、TIMP-1 mRNA表达。结果:与正常组比较,模型组COL-Iα1、TIMP-1mRNA的表达显著升高,差异有统计学意义(P<0.01);与模型组比较,下瘀血汤组、组分处方组COL-Iα1、TIMP-1mRNA的表达明显降低,差异有统计学意义(P<0.01);与组分处方组比较,下瘀血汤组COL-Iα1、TIMP-1mRNA表达明显降低,差异有统计学意义(P<0.01);相关性分析表明COL-Iα1、TIMP-1 mRNA的表达呈显著正相关。结论:下瘀血汤全药与组分处方可通过抑制TIMP-1 mRNA和COL-Iα1表达而发挥抗肝纤维化作用,且下瘀血汤全药处方作用优于组分处方。
Objective: To compare the effects of "Xiayuxue Decoction" and its componential prescription on expressions of COL-Iα1 and TIMP-1 in rats with immunologic hepatic fibrosis, and to explore the mechanisms of "Xiayuxue Decoction" and its eomponential prescription on anti-fibrotic liver tissue. Methods: Wistar rats were randomly divided into normal group and model established group. Rats in model established group were duplicated by porcine serum injection for 12 weeks, 0.5 ml for each rat, twice a week. After successful modeling, rats were randomly divided into model group, "Xiayuxue Decoction" group and componential prescription group. Rats in "Xiayuxue Decoction" group and componential prescription group were administered with 2.34 g/kg and 0. 648 g/kg medicine ( 10 ml/kg) per day by intragastric administration respectively for 4 weeks. Rats in normal group and model group were treated with 0.9% NaC1 of on the same volume. Liver tissue samples were achieved after sacrificing the rats at the end of 12 weeks, and the expression of COL-Ial and TIMP-1 mRNA were detected by Real-time PCR detection system. Results: Compared with normal group, expressions of COL-IcH and TIMP-1 mRNA in model group were increased significantly ( P 〈 0. 01 ) ; compared with model group, expressions of COL-Iα1 and TIMP-1 mRNA in "Xiayuxue Decoction" group and eomponential prescription group were decreased significantly ( P 〈 0.01 ) ; compared with componential prescription group, the expressions of COL-Iα1 and TIMP-1 mRNA in "Xiayuxue Decoction" group were decreased significantly (P 〈0.01 ). Correlation analysis showed that COL-Iα1 expression was correlative with TIMP-1 mRNA expression. Conclu- sion: "Xiayuxue Decoction" and its componential prescription can fight liver fibrosis by restraining expressions of TIMP-1 mR- NA and COL-Iα1, and the "Xiayuxue Decoction" has better clinical effect than its componential prescription.
