摘要
烟用酶制剂中糖化酶的活力是烟草工业生产中的重要指标。建立了3,5-二硝基水杨酸(DNS)测定糖化酶活力的方法。结果表明,在碱性条件下,糖化酶水解液与DNS反应产物的最大吸收波长为480 nm,线性范围为1.30~171.5 U/mL,加标回收率为99.8%~103.4%,RSD为0.7%~2.9%,检出限为1.58 U/mL,该方法可用于烟用酶制剂样品中糖化酶的测定,与标准方法相比,其准确性符合要求,且具有简便、快速的特点。
Glucoamylase activity of enzyme used in tobacco is an important index to tobacco industrial manufacturation.A method to determine glucoamylase activity by spectrophotometry with 3,5-dinitrosalicylic acid had been developed.The results indicated that under alkaline conditions,the maximum absorption wavelength of 3-amino-5-nitro-salicylic acid,which was formed by the reaction of glucose and 3,5-dinitrosalicylic acid,was 460nm,the linear relationship was in the range of 1.30~171.5 U/mL,the recovery rate was 99.8 %~103.4 %,and RSD was 0.7 %~2.9 %,the detection limit was 1.58 U /mL.Such method could be applied in the determination of glucoamylase activity in enzyme samples for tobacco use.Compared with standard method,it had the advantages including high accuracy,simple and rapid operation.
