PPAR-α/γ激动剂干预对代谢综合征大鼠肾功能的影响 被引量:1

摘要 目的探讨过氧化物酶体增殖物激活受体(PPAR)-α/γ激动剂干预对代谢综合征(MS)大鼠的糖脂代谢、肾功能的影响,为保护MS患者肾功能寻找新的思路和方法。方法高果糖饮食喂养SD大鼠构建MS大鼠模型,不同药物干预后,定期测定各组大鼠[模型对照组(MC)、非诺贝特组(FEN)、吡格列酮组(PIO)、非诺贝特+匹格列酮组(FEN+PIO)、空白对照组(NC)]的体质量、血压(SBP)、血糖(FBS)、血脂、肝肾功能等,分析各组上述指标间的差异及关系。结果①与NC大鼠相比较:MS大鼠SBP、FBS、尿酸(UA)、尿素氮(BUN)、肌苷(Cr)水平升高(P<0.01或P<0.05),体质量减轻(P<0.01)。②与MC大鼠相比:FEN组大鼠血清UA、Cr降低(P<0.01或P<0.05);③与FEN组大鼠比较:PIO组大鼠血清UA升高(P<0.05)。④与MC大鼠相比:FEN+PIO组大鼠血清UA、Cr降低(P<0.01或P<0.05)。结论 FEN单用或FEN+PIO干预可降低MS大鼠的血清UA、CR,对肾功能有保护作用。单用PIO干预,比单用FEN干预,使MS大鼠的UA升高。 Objective To investigate the impact of peroxisome proliferator-activated receptor-α/γ(PPAR-α/γ) agonist intervention on glucose and lipid metabolism,and renal function of rats with metabolic syndrome(MS) in order to find out new ideas and methods to protect the renal function in patients with MS.Methods The MS model of SD rat was induced by high-fructose diet.After the intervention of different drugs,the weight,systolic blood pressure(SBP),fasting plasma glucose(FBS),blood lipids,liver and kidney function of the rats in each group(the model control(MC) group,the fenofibrate(FEN) group,the pioglitazone(PIO) group,fenofibrate + pioglitazone(FEN + PIO) group,and the normal control(NC) group) were periodically determined.The differences and relationships between each group were analyzed.Results ①The levels of SBP,FBS,uric acid(UA),blood urea nitrogen(BUN) and carnine(CR) in MS rats were higher than those in NC group(P 〈 0.01 or P 〈 0.05),although the weight in MS rats was lower(P 〈 0.01).②The serum levels of UA and CR in the FEN group were lower compared with those in MC group(P 〈 0.01 or P 〈 0.05).③The serum level of UA in the PIO group was higher than that in the FEN group(P 〈 0.05).④The serum levels of UA and CR in the FEN + PIO group were lower than those in MC group(P 〈0.01 or P 〈0.05).Conclusion Rats treated with FEN only or with FEN + PIO could reduce the serum levels of UA and CR,which demonstrated they were able to protect the renal.The level of UA in PIO group was higher than that in FEN group.
