摘要 目的:探讨不同浓度的血管抑素对血管内皮细胞中VEGF表达的影响。方法:将体外培养的人脐静脉内皮细胞分为三组:对照组(空白组)、A组(16mg/L血管抑素组)、B组(32mg/L血管抑素组),通过MTT、RT-PCR、Western-Bolt等方法检测细胞内VEGF的表达情况。结果:随着血管抑素作用时间的延长和浓度的递增对人脐静脉内皮细胞的抑制作用也逐渐增强,且与对照组相比差异均有显著性意义(P<0.05)。结论:血管抑素通过抑制VEGF的表达以抑制人脐静脉内皮细胞增殖,从而抑制血管新生。 Objective To evaluate the inhibitory effects of angiostatin on VEGF for human umbilical vein endothelial cells (HUVEC). Methods Culturing HUVECs in vitro was divided i into three groups (group control:blank group;group A: 16 mg/L angiostatin; group B: 32 mg/L angiostatin) to observe the expression of the VEGF and the proliferation of HUVECs was examined by M]-F,RT-PCR and Western Bolt. Results angiostatin (16,and 32 rag/L) could significantly inhibit the growth of HUVECs after 24,48,and 72 hours (P〈0.05) in a dose-time-dependent manner. Conclusion Angiostatin can inhibit the proliferation of HUVECs and disturb the angiogenesis,because the AS can inhibit the expression of VEGF, so as to inhibit the growth and metastasis of CNV.
