NKG2D及其配体MICA/B在口腔鳞癌患者外周血NKT细胞中的表达及其意义 被引量:2

Expression and Significance of NKG2D and Its Ligand MICA/B of NKT Cells in the PBMC of Patients with Oral Squamous Cell Carcinoma

摘要 目的:探讨NKG2D及其配体在口腔鳞癌患者与正常人中外周血NKT的表达及其临床意义。方法:通过流式细胞术检测28例口腔鳞癌及25例正常人的NKT细胞计数,PCR检测NKG2D及其配体在不同组中NKT及配体MICA/B的表达情况,分析表达率与临床意义。结果:与正常组相比口腔鳞癌病人中的外周血中CD3+CD56+NKT的含量较低((1.74±0.15)个/μL),并且口腔鳞癌患者CD3+CD56+NKT细胞中NKG2D,MICA和MICB表达均下降,与对照组比较差异有统计学意义。结论:口腔鳞癌患者外周血中的NKT细胞数量和NKG2D及其配体MICA/B表达量相对下降,这可能影响NKT在肿瘤免疫中的作用,使口腔鳞癌细胞逃避免疫监视。他们可以作为口腔鳞癌免疫状态的参考指标,也为口腔鳞癌的治疗提供一种新的方向。 Objective:To research the significance and expression of NKG2D and its ligand MIA/B in NKT cells from the peripheral blood of normal group and patients with oral squamous cell carcinoma(OSCC).Methods:Flow cytometry was used to observe the NKT cells in the peripheral blood mononuclear cell of 28 oral squamous carcinoma patients and 25 healthy controls.The expression of NKG2D and its ligand MICA/B with polymerase chain reaction(PCR)in various persons.Results:In comparison with healthy volunteers,CD3+CD56+NKT in the peripheral blood of OSCC patients content was lower;the expression of NKG2D and its ligand MICA/MICB decreased in OSCC patients.And the difference was statistically significant.Conclusion:The low rate of the CD3+CD56+NKT in the peripheral blood and expression of NKG2D and MICA/B in the NKT of OSCC patients,which affect the role of NKT in tumour immunity,make tumour cells escape the immunological surveillance.They may be potential indicators of the oral tumour condition,and provide a new study direction for the treatment of oral squamous cell carcinoma.
