摘要
目的 :研究普罗帕酮与美托洛尔两药的相互作用 ,为临床合理联用提供依据 ;建立简便、准确的检测方法。方法 :采用日本Shimadzu公司的HPLC系统。结果 :①线性范围 普罗帕酮和美托洛尔回归方程分别为Y =- 1 12 42 +2 0 95 0X(r=0 9918) ,Y =- 0 0 0 6 4+0 2 72 6X(r =0 9978) ;②方法回收率 普罗帕酮和美托洛尔日内回收率分别为 (99 2± 5 6 ) %和 (10 0 0± 1 2 ) % ,日间回收率分别为 (97 8± 4 7) %和 (97.8± 4.1) ;③专一性 潘生丁、消心痛、维生素C、维生素E、氮化钾、地高辛、地塞米松等的出峰时间与两药的出峰时间均不重叠。结论 :可以用HPLC法简便、准确测定普罗帕酮与美托洛尔的血药浓度 ,同时处理样本 。
OBJECTIVE: A simple and rapid HPLC method for simutaneous determination of propafenone and netoprolol in plazma was established for studying intereactions of the two agents.METHODS: In the study a Shimadzu HPLC is adopted and propanolol is used as internal standard.RESULTS:① The linear limit:The regression equation for propafenone and metoprolo was Y=-1.1242+2.0950X(r=0.9918)and Y=-0.0064+0.2726X(r=0.9978)respectively;② The recovery:The recovery within day for propafenone and metoprolol was (99.2±5.6)% and(100.0±1.2)% respectively;the tecovery between day for propafenone and metoprolol was (97.8±4.7)% and (97.8±4.1)% repectively;③ The sepecificity: The peaks of dipyidamloe,nifedipine, vitamin C and E, KCl,digoxin,and dexamethasone did not overlaped each other.CONCLUSIONS:Plazma level propafenone and metoprolol can be simply, accurately and simutameously determined by the HPLC method established by the authors.
