糖脂平对2型糖尿病大鼠的治疗作用及机制 被引量:2

摘要 目的探讨糖脂平对2型糖尿病大鼠内质网应激和胰岛素抵抗的治疗作用机制。方法用小剂量链脲佐菌素(streptozotocin,STZ)腹腔注射联合高热量饲料喂养建立2型糖尿病大鼠模型。分为正常对照组(10只正常大鼠)、正常治疗组(10只正常大鼠,糖脂平治疗)、模型对照组(10只模型大鼠)和模型治疗组(10只模型大鼠,糖脂平治疗)。连续治疗8周后检测空腹血糖(fasting plasma glucose,FPG)、空腹血清胰岛素(fasting serum insulin,FINS),计算胰岛素敏感性指数(insulin action index,IAI),Western blot检测肝组织蛋白激酶R样内质网激酶(protein kinase R-likeER kinase,PERK)的磷酸化水平变化,荧光定量逆转录聚合酶链反应(fluorescent quantitative reverse transcription pol-ymerase chain reaction,FQ-RT-PCR)检测胰岛细胞C/EBP同源蛋白(C/EBP-homologous protein,CHOP)mRNA表达变化。结果模型治疗组在治疗8周后,与模型对照组比较,FPG显著降低,FINS和IAI显著升高;模型对照组磷酸化PERK及CHOP的mRNA的表达明显高于其他3组。结论糖脂平能有效降低2型糖尿病模型大鼠血糖,增加组织对胰岛素敏感性,可能机制是糖脂平通过减少PERK磷酸化水平和CHOP表达缓解2型糖尿病大鼠的内质网应激。 Objective To investigate the effects and mechanism of Tangzhiping on reducing endoplasmic reticulum stress(ERS) and enhancing insulin sensitivity in a rat model of type 2 diabetes.Methods Type 2 diabetes rat models were induced by peritoneal STZ injection and high fatty and high sugar diet feeding.Rats were divided into normal control group(10 normal rats),normal-treatment group(10 normal rats,treated by Tangzhiping),model control group(10 model rats),and model-treatment group(10 model rats,treated by Tangzhiping).Afer 8 weeks,the fasting plasma glucose(FPG) and the fasting serum insulin(FINS) were detected,the insulin sensitivity index was calculated,the expressions of protein kinase R-like ER kinase(PERK) and C/EBP-homologous protein(CHOP) were detected by Western blot and fluorescent quantitative reverse transcription polymerase chain reaction(FQ-RT-PCR) respectively.Results After Tangzhiping treatment for 8 weeks,the FPG of the model-treatment group was significantly lower,and the FINS and the IAI levels were significantly higher of the model-treatment group than those of the model control group.The expression of phosphorylated PERK protein and CHOP mRNA of the model control group were significantly higher compared with the other 3 groups.Conclusion Tangzhiping can effectively reduce blood glucose and increase insulin sensitivity in the rats with type 2 diabetes.The possible mechanism is that Tangzhiping treatment can relieve ERS through reducing the expression of phosphorylated PERK and CHOP mRNA.
