摘要
观察内皮素A受体拮抗剂BQ -610对脓毒症大鼠呼吸力学和动脉血气的影响。方法 30只雄性SD大鼠随机分成正常对照组、对照组、治疗A组、治疗B组和预防用药组。通过持续静脉泵注大肠杆菌内毒素复制大鼠脓毒症模型 ,分别监测治疗组治疗前后和其它组不同时相的气道阻力 (Raw)、肺动态顺应性 (Cdyn)和动脉血气的变化。结果 对照组大鼠Raw、PaCO2 明显上升 ,Cdyn、PaO2 明显下降 ,与正常对照组同时相相同指标比具有非常显著性差异 (P <0 0 1) ;BQ -610预防用药组上述指标与正常组相比无显著性差异 (P >0 0 5) ,与对照组相同时相的上述指标相比具有非常显著性差异 (P <0 0 1) ;治疗A组治疗前后上述指标有明显差异 (P <0 0 5) ;治疗B组治疗前后上述指标早期无明显差异 (P >0 0 5) ,但后期治疗B组治疗前后上述指标有较明显的差异 (P <0 0 5) ;A组和B组之间同时相上述指标相比也有显著性差异 (P <0 0 5)。结论 ET -1和ETAR参与了脓毒症和脓毒性休克的病理生理过程 ;在脓毒症时 ,内皮素A受体拮抗剂BQ -610可降低气道阻力 ,改善动态肺顺应性和肺内气体交换。
Objective To investigate the effect of BQ-610 on respiratory dynamics in the sepsis rats:Methods Thirty male SD rats were divided into normal,control,prevention,treatment A and B group,The changes of airways resistance(Raw),dynamic pulmonary compliance (Cdyn),Arterial oxygen tension (PaO 2)and Arterial carbon dioxide(PaCO 2)were measured in every group Results There are significant differences between the prevention group/normal group and control group in Raw,Cdyn,PaO 2 and PaCO 2 ( P <0 01) Raw increased and Cdyn,PaO 2 decrease in control groups,PaCO 2 decrease too in prophase But the PaCO 2 increased in anaphase( P >0 05) PaO 2,PaCO 2 had no significant improved in treatment A group;but had a significant improved in treatment B in prophase Conclusions selective ETAR antagonist BQ-610 may significantly decrease Raw,improve Cdyn,improve pulmonary function in sepsis and septic shock rats and had concentration-dependent manner ET-1 and ETAR suggest an important role in the mechanisms and pathogenesis of sepsis [
