摘要
目的探索从中华眼镜蛇(Chinese cobra,Naja naja atra)蛇毒中一次性快速分离提纯具有抗补体活性及溶血活性的中华眼镜蛇蛇毒因子子质量测定,并鉴定其活性与毒性。结果本法CCVF得率高于2%,电泳图谱呈单一区带,由3个亚基组成,相对分子质量为145 800,抗补体及溶血活性强,毒性低。结论本法操作简单,时间短,纯度高,为一实用高效快速提纯方法。
Objective To isolate and purify the Chinese cobra venom factor (CCVF) with strong anti-complementary activity from the Chinese cobra venom quickly. Methods CCVF was fractionsted by ion-exchange chromatography with PEAE-Saphadex A-50 column. By polyacrylamide slab gel electrophoresis, CCVF was identified and the molecular weight was estimated. The complement inhibition activity and the toxicity were tested. Results The extraction rate of CCVF was more than 2%, which presented a homogenous strip on the electrophoresis. With strong anti-complementary activity and low toxicity, CCVF consists of 3 subunites with total molecular weight of 145 800. Conclusion It was an efficient and simple method of CCVF fractionation.
