髂股动脉粥样硬化闭塞症家兔模型的制作 被引量:14

Establishment of iliofemoral atherosclerosis obliterans model in rabbits

摘要 目的 探讨动脉粥样硬化闭塞症 (atherosclerosisobliterans ,ASO)动物模型的制作方法。方法 实验组、对照组各 1 0只新西兰家兔 ,利用机械损伤、免疫损伤和高脂高胆固醇饲养 2 0周 ,观察模型制作前后血脂 (TC)、胆固醇 (TG)、内皮素 (ET)、一氧化氮 (NO)水平及股动脉造影和病理学变化。结果 制作前TC、TG、ET和NO分别为 ( 0 .89± 0 .0 9)mmol/L、( 0 .99± 0 .1 5)mmol/L、( 99.1 3± 1 6.51 )ng/L、( 6.2 6± 0 .80 ) μmol/L ,制作后分别为 ( 8.95± 1 .68)mmol/L、( 8.38± 1 .0 8)mmol/L、( 1 835.81± 2 0 3.62 )ng/L、( 2 .33± 0 .55) μmol/L。前后比较差异有非常显著性 ( P <0 .0 1 ) ;制作后实验组动脉造影显示髂股动脉高度狭窄 ,病理切片显示典型的ASO变化 ,对照组无异常变化。 Objective To investigate the establishment of the iliofemoral atherosclerosis obliterans (ASO) model in rabbits.Methods Twenty Newzland rabbits were randomly divided into experimental group and control group equally.The experimental group was raised by the fodder rich in glycerid and cholesterol for 20 weeks with the mechanic injuries and immune injuries,while the control group was raised by normal fodder.Before and after the development of the models the levels of serum glyceride,serum cholesterol,plasma ET and plasma NO were determined,iliofemoral vessels were examined by arteriography,and all iliofemoral arteries were examined pathologically at end.Results The levels of total serum glyceride,serum cholesterol,plasma ET and NO were (0.89±0.09) mmol/L,(0.99±0.15) mmol/L,(99.13±16.51) ng/L,(6.26±0.80) μmol/L before the establishment of the models,while the levels of post were (8.95±1.68) mmol/L, (8.38±1.08) mmol/L, (1 835.81±203.62) ng/L,(2.33±0.55) μmol/L respectively (P<0.01).The iliofemoral artery stricture and ASO pathological signs analogized to human ASO were found in arteriography and pathological examination.Conclusion This method can induce ASO rabbit models successfully.
