Study on the correlation between HOXA11 and polycystic ovary syndrome

摘要 目的:探讨HOXA11基因在卵巢颗粒细胞内的异常表达与PCOS的相关性。方法:采用免疫荧光法、RT-PCR法、Western-blot法检测颗粒细胞中HOXA11的表达水平。结果:①在PCOS组中,镜下细胞内见明显的红色荧光主要定位于细胞核,细胞液中可见少许微弱红色荧光;对照组中,细胞核和细胞液中可见更强的红色荧光。PCOS组与对照组比较,细胞核和细胞液中HOXA11的表达都明显减少(P<0.05)。②PCOS组HOXA11 mRNA的表达水平较对照组低(P<0.05)。③Western-blot结果显示,PCOS组与对照组的颗粒细胞中均有HOXA11蛋白表达。PCOS组表达较弱,对照组表达较强(P<0.05)。免疫荧光、RT-PCR、Western-blot均提示PCOS组颗粒细胞中HOXA11表达水平较对照组低(P<0.05)。结论:HOXA11在PCOS组的颗粒细胞中表达降低,提示其可能在PCOS发生发展过程中起着重要作用。 Objective: To explore the correlation between abnormal expression of HOXAll gene in ovarian granulosa cells and polycystic ovary syndrome (PCOS) . Methods: Immunofluorescence method, RT -PCR, and Western blot were used to detect the expres- sion level of HOXA11 gene in ovarian granulosa cells. Results: In PCOS group, obvious red fluorescence mainly located in cell nucleus, there was a little weak fluorescence in cytoplasm; in control group, stronger red fluorescence could be observed in cell nucleus and cyto- plasm. The expression levels of HOXA11 in cell nucleus and cytoplasm decreased significantly in PCOS group and control group (P 〈 0. 05 ) . The expression level of HOXA11 mRNA in PCOS group was significantly lower than that in control group ( P 〈 0. 05 ) . The results of western blot showed that HOXA11 protein expressed in granulosa cells of PCOS group and control group, the expression in PCOS group was weaker than that in control group ( P 〈 0.05 ) . The results of immunofluorescence method, RT - PCR, and Western blot indicated that the expression level of HOXA11 in granulosa cells of PCOS group was significantly lower than that in granulosa cells of control group ( P 〈 0. 05 ) . Conclusion : The expression of HOXA11 in granulosa cells of PCOS group decreased, which indicates that HOXA11 plays an im- portant role in the occurrence and development of PCOS.
