摘要
目的探讨莪术醇对人鼻咽癌CNE-2细胞的凋亡作用及其分子机制。方法以不同浓度的莪术醇处理体外培养的人鼻咽癌CNE-2细胞。采用流式细胞仪检测细胞凋亡率,用western blot检测核因子NF-κB、B细胞淋巴瘤/白血病-2基因(Bcl-2)及血管内皮生成因子(VEGF)的表达。结果流式细胞仪检测结果显示莪术醇处理组与阴性对照组CNE-2细胞凋亡率有显著性差异(P<0.01),且呈量效关系;Western blot检测显示NF-κB、Bcl-2和VEGF的表达水平随莪术醇剂量的增大而下降(P<0.01)。结论莪术醇能够诱导鼻咽癌CNE-2细胞的凋亡,其诱导细胞凋亡的机制可能是通过抑制NF-κB表达,从而影响了Bcl-2转录激活;此外莪术醇还可抑制VEGF的表达。
Objective To investigate the molecular mechanism of cureumol in nasopharyngeal carcinoma cell line CNE - 2 . Methods CNE -2 cells were treated with curcumol at different concentrations. The apoptosis was analyzed by flow cytometry( FCM ). The expression of nuclear factor - κB ( NF - κB), B - cell lymphoma/leukemia - 2 gene ( Bcl - 2) and vascular endothe- lial growth factor (VEGF) were detected by western blot. Results Apoptosis induced by curcumol in CNE -2 cells was in a dose - dependent manner. The expression of NF - κB, Bcl - 2 and YEGF was down - regulated with the increasing concentration of curcumol. Conclusion Curcumol is capable of inducing apoptosis of CNE - 2 cells in vitro significantly. The mechanism may be related to the inhibition of Bcl - 2 transcription through down - regulation of the NF - KB protein level. In addition curcumol signif- icantly inhibits the VEGF expression in CNE- 2 cells.
